Objectives:The purpose of the study was to detect the expressions of SDF-1 and CXCR4 in colorectal adenocarcinoma,to evaluate their roles in carcinogenesis,invasion and metastasis as well as their clinical significances,to provide theoretical foundation for early finding and intervention and new target for targeted therapy.Methods:1.The expressions of SDF-1 and its receptor CXCR4 were detected by SP immunohistochemical method in 60 cases of colorectal adenocarcinoma,23 cases of colorectal adenoma and 22 cases of normal colorectal mucosa.2.The expressions of SDF-1mRNA and CXCR4mRNA were detected by RT-PCR method in 30 cases of colorectal adenocarcinoma,15 cases of normal colorectal mucosa.Results:1.the results of immunochemistry The differences of SDF-1 and CXCR4 proteins positive expression among the colorectal adenocarcinoma,colorectal adenoma and the normal colorectal mucosa are significant(P<0.05,P<0.05),and the positive expressions of SDF-1 and CXCR4 in colorectal cancer were the highest in the three groups(P<0.05,P<0.05);Stronger expressions of SDF-1 and CXCR4 proteins were observed in colorectal adenoma,compared with those in normal tissues(P<0.05,P<0.05). The positive expressions of SDF-1 and CXCR4 were closely correlated with the depth of tumor infiltration and metastasis of lymph node:the positive expressions of SDF-1 and CXCR4 in invasion to chorion were obviously higher than those invasion to mucosa or muscular layer(P<0.05,P<0.05);the positive expressions of SDF-1 and CXCR4 in colorectal adenocarcinoma with lymphnode metastasis was obviously higher than those without lymphnode metastasis(P<0.01,P<0.01).But they were not related to gender and age of patients and differentiation of cancer(P>0.05).2.the resultjs of RT-PCR The expressional quantities of SDF-1mRNA,CXCR4mRNA in the colorectal adenocarcinoma were obviously higher than those in the normal colorectal mucosa(P<0.01,P<0.01),and were closely correlated with depth of tumor infiltration and metastasis of lymph node:the expressional quantities of SDF-1mRNA,CXCR4mRNA in invasion to chorion were obviously higher than those invasion to mucosa or muscular layer(P<0.05,P<0.05);the expressional quantities of SDF-1mRNA,CXCR4mRNA in colorectal adenocarcinoma with lymphnode metastasis were obviously higher than those without lymphnode metastasis(P<0.01,P<0.01). 3.Correlation analysis(l)The positive expressions of SDF-1 and CXCR4 proteins are positively correlated in the 60 cases of colorectal adenocarcinoma(r=0.436,P<0.01).(2) The expressional quantity of SDF-1mRNA and CXCR4mRNA are highly positively correlated in the 30 cases of colorectal adenocarcinoma(r=0.949,P<0.01).(3)The positive expression of SDF-1 protein and expressional quantity of SDF-1mRNA are positively correlated in the 30 cases of colorectal adenocarcinoma(r=0.480,P<0.01).(4) The positive expression of CXCR4 protein and expressional quantity of CXCR4mRNA are positively correlated in the 30 cases of colorectal adenocarcinoma(r=0.588, P<0.01).Conlusion:1.SDF-1 and its receptor CXCR4 may play an important role in tumorigenesis progression,infiltration and metastasis of colorectal adenocarcinoma.2.To detect the expressions of SDF-1 and CXCR4 may provide guidance for the early finding,predicting prognosis and targeted therapy of colorectal adenocarcinoma.
