Objectives:The pupose of the study was to detect the expressions of NF-κBp65,ⅠκBa and HPA in colorectal adenocarcinoma,to evaluate their roles in carcinogenesis, invasion and metastasis as well as their correlation in colorectal adenocarcimoma,to provide theoretical foundation for early finding and intervention and new target for targeted therapy.Methods:1.The expressions of NF-κBp65 and HPA were detected by SP immunohistochemical method in 60 cases of colorectal adenocarcinoma,23 cases of colorectal adenoma and 26 cases of normal colorectal mucosa.2.The expressions ofⅠκBamRNA and HPAmRNA were detected by RT-PCR method in 40 cases of colorectal adenocarcinoma,15 cases of normal colorectal mucosa.Results:1.the results of immunochemistry The positive expression of NF-κBp65 and HPA in the colorectal adenocarcinoma was obviously higher than those in the colorectal adenoma and the normal colorectal mucosa(P<0.05),and was closely correlated with depth of tumor infiltration and metastasis of lymph node:the potive expression of NF-κBp65 and HPA in invasion to chorion was obviously higher than those invasion to the muscular layer(P<0.05,P<0.05);the positive expression of NF-κBp65 and HPA in colorectal adenocarcinoma with lymphnode metastasis was obviously higher than those without lymphnode metastasis(P<0.01,P<0.05).2. the result of RT-PCR The expressional quantity ofⅠκBamRNA,HPAmRNA in the colorectal adenocarcinoma was obviously higher than those in the normal colorectal mucosa(t=2.644,2.226,P<0.05,P<0.05),and was closely correlated with depth of tumor infiltration and metastasis of lymph node:the expressional quantity ofⅠκBamRNA,HPAmRNA in invasion to chorion was obviously higher than those invasion to the muscular layer(t=4.093,3.380,P<0.01,P<0.01); the expressional quantity ofⅠκBamRNA,HPAmRNA in colorectal adenocarcinoma with lymphnode metastasis was obviously higher than those without lymphnode metastasis(t=4.576,4.366,P<0.01,P<0.01).3.Correlation analysis(1) The positive expression of NF-κBp65 and expressional quantity ofⅠκBmRNA is highly positive correlated in the 40 cases of colorectal adenocarcinoma(r=0.732,P<0.01)(2)The positive expressions of NF-κBp65 and HPA is positive correlated in the 60 cases of colorectal adenocarcinoma(r=0.517,P<0.01)(3)The expressional quantity ofⅠκBmRNA and HPAmRNA is positive correlated in the 40 cases of colorectal adenocarcinoma(r=0.408,P<0.01)Conlusion:1.NF-κB and HPA are correlative factors of colorectal adenocarcinoma,NF-κB and HPA may play an important role in tumorigenesis progression,infiltration and metastasis of colorectal adenocarcinoma.2.NF-κB may promote the infiltration and metastasis in colorectal adenocarcinoma by up-regulating the expression of HPA.3.To detect the expressions of NF-κB and HPA may provide guidance for the early finding,predicting prognosis and targeted therapy of colorectal adenocarcinoma.
