ObjectiveTo study the expression and significance of P53 and NF-κB(Nuclear Factor Kappa B)P65 in gastric adenocarcinoma, tissues beside tumor, chronic atrophic gastritis and normal gastric mucosa tissues respectively, through analysis of differences in expressions and relationship between the four mucosa tissues of clinicopathological factors, roles of P53 and NF-κBP65 in the occurrence, devel opment and metastasis of gastric adenocarcinoma are to be explored,which will prefer a experimental evidences on the effect on anti-tumor.MethodologyS-P immunohistochemical technique was adopted to test P53 and NF-κBP65 proteins of 100 gastric adenocarcinoma and 40 tissues beside tumor chronic atrophic gastritis and normal gastric mucosa tissues respectively, and the factor of age,gender,size,differentiated degree,lympho node metastasis and depth of invasion are all analysis in aggregate of gastric adenocarcinoma. Statistical analysis was conducted with the data using Wilcoxon Kruskal-Wallis rank-sum test and Spearman rank correlation analytical method.Results1.Expression of P53 protein was appearing as brown granules is located in the cell nucleus.Expression rates in gastric adenocarcinoma, tissues beside tumor, chronic atrophic gastritis and normal gastric mucosa tissues are 56.0%, 22.5%,7.5% and 2.5% respectively, the differences of which are statistically significant(2χ=55.72, P< 0.01). Expression of P53 has not been found to be affected by patients’ age, gender,size,differentiated degree and depth of invasion,but correlation with whether metastasis lympho node((2χ=5.45,P<0.05).2.Expression of NF-κBP65 protein was appearing as brown granules is located in the cell nucleus and cytoplasm. Expression rates in gastric adenocarcinoma,chronic atrophic gastritis, tissues beside tumor and normal gastric mucosa tissues are 71.0%, 15.0% and 12.5% and 5.0% respectively, the differences of which are statistically significant. The differences between four groups are statistically significant(2χ=84.57, P < 0.01). Positive rates of NF-κBP65 in gastric adenocarcinoma histological grades of highly, the differences of which are statistically significant and positively correlated to histological grades of tumors(2χ=7.47,P<0.05),and correlation with whether metastasis lympho node((2χ=5.70,P<0.05).Expression of NF-κBP65 has not been found to be related to age, gender,size and depth of invasion and so on.3.Correlation of expressions of P53 and NF-κB P65 in gastric adenocarcinoma of 100 cases with gastric adenocarcinoma,the correlation coefficient is 0.334, P<0.05.Conclusion1.A sharp rise of NF-κBP65 and P53 protein expression in gastric adenocarcinoma tissue was found and positively related to histological grade of tumors, indicating that the stronger proliferation activity is, the higher grade malignancy is.2.A sharp rise of NF-κBP65 and P53 protein expression in gastric adenocarcinoma tissue was found and closely related to lymph node metastasis, indicating that ability to promote lymphatic hyperplasia more strong. NF-κBP65 is positively related to degrees of differentiation.3. Two factors of P53 and NF-κB in the development of gastric adenocarcinoma were positively correlate.4.Correlation between expressions of P53 and NF-κBP65 in gastric adenocarcinoma has been found.They can be used as joint detection factor.
