Objective:This article aims to make the model of lower limb i schemia in diabetic rats and tostudy the therapeutic effect of Xia oke tongbi granule on diabetic lower limb ischemia rats. By obser ving the expressive effects of Xiaoke tongbi granule on the PI3K、A kt mRNA in diabetic lower limb ischemia rats, we can study prelim inary on the effects of Xiaoke tongbi granule on the PI3K-Akt path way in diabetic lower limb ischemia rats。Methods:We prepare 100 clean SD rats for the experiment. Then we randomly take 10 rats as blank group which are fed with ordina ry forage. Meanwhile the rest of the rats are fed with forage high in fat. After one month, the rats fed with forage high in fat ar e given an abdominal cavity injection of STZ (45mg/kg) in the fas ting state for the model. Then we take the rats whose blood gluco se more than 16.7mmol/L into the experiment. Finally, these select ed rats are operated with arteriovenous ligation in the left leg femoral to make the model of diabetic lower limb ischemia. The su ccessfully established experimental rats models were randomly divi ded as model group, High, middle and small dose groups for Xiaoke tongbi granule, Western Medicine (cilostazol) group with 10 rats i n each group. Meanwhile they are lavaged. The normal group and th e model group are fed with physiological saline in 10ml/kg/d. High dose treatment groups are fed with Xiaoke tongbi granule in 1.875 g/kg/d, middle dose group are fed with Xiaoke tongbi granule in 1. 35g/kg/d, small dose group are fed with Xiaoke tongbi granule in 0. 625g/kg/d, western medicine group are fed in 45mg/kg/d. After 6 we eks, the experimental rats were killed. Then we observe the level of blood glucose, body weight and serum insulin levels. Meanwhile we also need to observe the changing of nerve pathological ischem ic areas in rats and use PCR technique to detect the expression of PI3K, Akt mRNA in muscle ischemia regionsResult:When we observe the neural morphology of the ischemic area for rats with light microscope, we can find that model group rats present pathological ischemic injury, the body weight of rats significantly decrease and blood glucose increase significantly (P<0.01); The effect of Xiaoke tongbi granule can slow down declining trend of the weight of model rats, reduce the level of blood glucose, decrease the level of serum insulin and improve insulin resistance in rats. Simultaneously the middle dose group is better than the High and small dose groups (P<0.05). Compared with the blank group and model group, all of treatment groups increased expression of PI3K,、Akt mRNA. However cilostazol western medicine treatment group increased significantly (P<0.01). For the increasement in the expression of PI3K,-Akt mRNA, middle dose’group for Chinese medicine is better than small and middle dose groups.Conclusion:The effect of Xiaoke tongbi granule can improve the general state of diabetic lower limb ischemia rats, improve the level of the blood glucose and insulin resistance. They can increase the expression of the PI3K、Akt mRNA in ischemic muscles to upregulate the PI3K-Akt pathway, promote the formation of new blood vessels in ischemic nerve and surrounding tissue and inhibit diabetic peripheral neuropathy.
