Objectives The purpose of this study is to compare the concentration of MIP-1α andTNF-α in gingival crevicular fluid of chronic periodontitis before and after periodontalinitial therapy by Enzyme-linked immunosorbent assay, analyze MIP-1α and TNF-αlevels before and after periodontal initial therapy and the relationship between MIP-1αand TNF-α and clinical parameters,explore whether MIP-1α and TNF-α could be anobjective indicator for monitoring the effects of periodontal initial therapy.Methods Research subjects: Patients with chronic periodontitis were selected sinceDecember2012to December2013, from the Department of Stomatology of Coal GeneralHospital. The study consisted20patients with chronic periodontitis and20healthycontrols from out-patient clinic.Including criteria: no fillings, caries or larger calculus inthe neck of the measuring tooth, with gingival index>1, periodontal pocket PD≥4mm,attachment loss≥3mm, different degree of alveolar bone absorption were found in X-ray.Experimental process: screening newly diagnosed patients met the inclusion criteria, thecommunication, written informed consent. Patients with chronic periodontitis shootingpanoramic radiographs, according to the need, where appropriate, plus photos apicalslices. One week after referral diagnosed periodontal health group based periodontalexamination records newly diagnosed with sterile instruments to remove manual scalingsubgingival plaque on teeth observed by medical sterilization lap across the wet, wipe drysterile cotton dental and record the GI, BI, PD, AL clinical indicators after face gently drywith an air gun test tooth surface and surrounding gum mucosa, GCF samples collectedpreservation. Then underwent periodontal therapy (full mouthsubgingival scaling technique and once completed subgingival scaling), after basictreatment is completed with3%hydrogen peroxide solution rinse periodontal,1%iodineglycerin and finally oral hygiene. Before treatment of chronicperiodontitis group and the healthy control group in the gingival index, bleeding index(BI), probing depth, attachment loss to compare aspects have a significant difference (P <0.01), with statistical significance; underlying chronic periodontitis patients aftertreatment, clinical indicators measured tooth has significant difference was statisticallysignificant (P<0.01). MIP-1α expression levels in patients with chronic periodontitis GCFwas significantly higher than in the healthy group, GCF levels, there is a significantdifference (P<0.01); GCF after periodontal treatment in the MIP-1α expression level wassignificantly lower than before treatment, a statistically significant (P<0.01). Beforetreatment, the patients with chronic periodontitis in GCF very significantly higher thannormal levels of TNF-α group (P<0.01), significantly lower TNF-α before and afterperiodontal treatment of chronic periodontitis contrast, treatment group prior to thetreatment group (P<0.01).Conclusions The clinical parameters reduced after foundation treatment, the differenceswere statistically significant.Indicates that foundation treatment can be effective in thetreatment of periodontal disease. Result the MIP-1α and TNF-α levels were higher in thechronic periodontitis group than in the healthy group, after non-surgical treatment cansignificantly reduce the concentration of MIP-1α and TNF-α. Indicates that MIP-1α andTNF-α have a certain relationship with periodontitis.Patients with chronic periodontitis inGCF MIP-1α, TNF-α level changes for understanding changes in condition, observing theefficacy and prognosis have some clinical value.
