Study Of Effect Of Ointment Rubbing On Mediators Of Inflammation Of Rats With Knee Osteoarthritis

Objective:To investigate the effect of Chenyuan Ointment rubbing on the cartilage tissue morphology, the mesochondrium and the levels of NO and PGE2in serum of rats with knee osteoarthritis(KOA), as well as to explore if there is a repair effect on cartilage, and to explore whether it has anti-inflammatory effects. Methods:52SD rats were randomly divided into4groups:Normal group, Model group, Chenyuan cream group and Calligraphy control group. KOA model was induced by closed-plaster method.8weeks after fixation, Normal group and Model group without intervention, rats in Chenyuan cream group and Calligraphy control group were given Chenyuan cream with manipulation and Vaseline with manipulation respectively once a day for8weeks. Then detect concentration levels of NO and PGE2in serum. Cut the left knee femoral condyle, then make pathological section by using HE staining and immunohistochemistry to evaluate the morphology score of cartilage, as well, to detect the expression of collagen type I and type II. Results:Compared with Model group, although the two treatment groups all showed lower expression level of collagen type I and Mankin score(P<0.05), Chenyuan cream group showed higher expression level of collagen type II (P<0.05), while Calligraphy control group had no significant difference(P>0.05). Moreover, both Chenyuan cream group and Calligraphy control group showed lower serum levels of NO and PGE2than Model group (P<0.01), while Chenyuan cream group showed the lower serum levels than Calligraphy control group (P<0.05). Compared with Model group, lower limb circumference and activity of knee joint of rats in two treatment groups were significantly improved after the treatment (P<0.01), while Chenyuan cream group improved greatly (P<0.05). Conclusion:Compared with vaseline with manipulative treatment, Chenyuan Ointment rubbing could enhance the expression of collagen type II and maintain the normal collagen phenol type, reduce the concentration level of NO and PGE2in serum more significantly, and improve muscle strength of lower limbs and activity of knee joint better, which could play a role in repairing articular cartilage and alleviating inflammatory reaction to improve the function of knee joint better.
