Objective:Combined with modern medicine on tourette syndrome(TS) neurotransmitter imbalance hypothesis,we had experiment to know that whether Dingchou granule has the regulation of amino acid neurotransmitter role in order to further elucidate the mechanism of action of Dingchou granule in the treatment of Tourette syndrome.Methods:60mice were randomly dived into6groups,namely,the normal control group,model group,high dose Dingchou granule group(treated by high dose dingchou granule),median dose Dingchou granule group(treated by median dose dingchou granule),low dose dingchou granule group(treated by low dose dingchou granule),positive control group(treated by Haloperidol).There were12animals in each group.We used the method of intraperitoneal injection of IDPN to induced TS animal model and HPLC for determination of glutamic acid, aspartic acid, butyric acid amino acid in plasma and brain tissue of mice.Results:1.The brain tissue levels of GLU,ASP in model group was increased and the levels of GABA was decreased,compared with that of the normal control group(P<0.01).2.The plasma levels of GABA in model group was decreased comared with normal control group(P<0.01).The results of GLU,ASP were not founded in the plasma.3.Compared with the model group,the brain tissue levels of GLU,ASP,GABA in the all dingchou groups had statistical significance(P<0.01), and there wao no statistical significance comapred with the positive control group (P>0.05)4.The brain tissue levels of GABA in all dingchou groups had statistical significance compared with the model group(P<0.01).There was statistical significance compared the high pose dingchou group with the positive group and no statistical significance compared median pose,low pose dingchou group with the positive group (P>0.05)Conclusion:1.It has Imbalance of amino acid neurotransmitters in brain tissiue and plasma of mice that was induced by IDPN.2.Dingchou granule can increase the content of GABA in brain tissue and plasma of mice model and decrease the content of GLU,ASP in brain tissue of mice model.3.Mechanism of action of dingchou granule in the treatment of TS may have relationship with increasing the content Of excitatory amino acids and decreaseing the level of nhibitory amino acids of the brain.
