Clinical Research Objective:To evaluate efficacy of Dingchou Granule on TS due to obstruction of collateral caused by wind-phlegm.Methods:Sixty patients consistence with diagnostic criteria were equally divided into the experimental group and the control group in a random,single-blinded,positive and parallel control way.The experimental group was administered with Dingchou Granule,and the control group was administered with Tiapride.12 weeks was a course of treatment.We observed the tic category,frequency, intensity and the improvement of effect on life or behavior in both groups before and after the treatment according to the Yale global tic severity scale(YGTSS).Results:Compared with the control group,there was a significant difference(P<0.05) in the experimental group in improving the tic category,frequency and the therapeutic effect.Compared with the control group,there was no significant difference(P>0.05) in the experimental group in improving the tic intensity and the effect on life or behavior.Conclusion:Dingchou Granule is effective on TS due to obstruction of collateral caused by wind-phlegm.Empirical Study Objective:To observe the pharmacodynamic action of this compound formula on TS in mice models caused by IDPN.Methods:In a random, double-blinded,positive and parallel control way,mice were divided into six group:the blank group,the model group,the Dingchou Granule group(low dose,moderate dose,high dose) and the haloperidol group,12 mice in each group.After being received peritoneal injection for 7days and from the day when the peritoneal injection was carried out,the experimental group was received intragastric administration with Dingchou Granule and the control group with haloperidol for 3 weeks,by observing stereotyped behavior and motor behavior in mice and determining the encephalon monoamine neurotransmitter and its metabolic products in mice with HPLC.Results:There were two aspects in ethology: 1.Dingchou Granule could significantly improve stereotyped behavior in mice,compared with the model group(P<0.01);and compared with the positive control group (P>0.05).2.Dingchou Granule could significantly improve motor behavior in mice,and compared with the model group(P<0.01);and compared with the positive control group (P<0.01).There were five aspects in monoamine neurotransmitter and its metabolic products:1.Dingchou Granule(low dose,moderate dose and high dose) could decrease the DA contents in mice.Compared with the model group(P<0.01);compared with the haloperidol group(P<0.01).2.Dingchou Granule in high dose could decrease NE contents in mice’s brain.Compared with the model group(P<0.05).3.Haloperidol could increase the HVA contents in mice’s brain.Compared with the model group(P<0.05).4.Haloperidol could increase the 5-HIAA contents in mice’s brain.Compared with the model group (P<0.01).5.There was no significant difference(P>0.05) on 5-HT and DOPAC among the Dingchou Granule group,the haloperidol group and the model group.Conclusion: Improving the tic behavior by decreasing the DA and NE contents may be the mechanism of the compound formula.
