Background:Tourette Syndrome in Children is closely related to neurotransmitter imbalance and oxidative stress response injury.Acupuncture therapy is one of the important effective therapies for it.Based on theoretical research,this study explores the mechanism of acupuncture therapy by observing the therapeutic effect of wind calming and spasmolytic therapy on model rats and its influence on neurotransmitter and oxidative stress indexes.Objective:On the basis of theoretical research,the behavioral changes of model rats,the expressions of neurotransmitters,oxides,antioxidants in serum and receptor signaling proteins in brain tissue were observed,and the mechanism of acupuncture for calming the wind and relieving spasm in treating Tourette's Syndrome was discussed,providing laboratory basis for clinical application of acupuncture in treating this disease.Methods:Fifty SD rats were divided into cages,fed adaptively,and given regular diet for 5 days.They were randomly divided into 5 groups:control group,model group,tiapride group,acupuncture group 1 and acupuncture group 2,each with 10 rats.Except for the control group,the other four groups of rats were injected intraperitoneally with selective 5-HT(2A/2C)receptor agonist 1-(2,5-Di Methoxy-4-Iodopheny L)-2-Amin Opropane(DOI)1 mg/kg to prepare a rat model of Tourette's syndrome,which was established for 3 weeks.When rats involuntarily show persistent rigid movements such as head and shoulder twitching and shaking,the modeling is successful.During the administration,tiapride group was given 15 mg kg-1 intragastric administration,control group and model group were given the same volume of normal saline intragastric administration,group 1 was acupuncture Pishu and Shenshu points,and group 2 was acupuncture Dazhui and Shenzhu points.Operation method:The rats were fixed.After local shaving of the acupoints,Huatuo brand acupuncture needles of 25mm × 25mm were used to directly prick 4-5 mm.The needle is retained for 10min,and is inserted once every 3min,5 days a week and once a day for a total of 21 days.Behavioral observation was carried out on rats.Enzyme-linked immunosorbent assay(ELISA)was used to detect the content of neurotransmitters in serum.Thiobarbituric acid(TBA method),superoxide dismutase WST-1 method and trace enzyme labeling method were used to detect the changes of oxide(MDA)and antioxidant(SOD,GSH).Western Blot technique was used to detect the expression of D1,D2 and NMDA receptors in brain tissue of TS rats.Results:1.Effect of Acupuncture on Behavior of Rats with Tourette SyndromeCompared with the control group,the sports score in the model group was significantly higher(respectively 24.66±4.14 scores;0±0 scores),with statistical significance(P<0.01),indicating that the model was successfully built.Compared with model group,acupuncture group 1 and acupuncture group 2 had significantly lower motor scores(respectively 18.66±5.09 scores;13.17±4.51 scores),indicating that acupuncture treatment is effective,and the effect of acupuncture group 2 is more obvious,with statistical significance(P<0.01).Results:Acupuncture at Dazhui and Shenzhu points is more effective than Pishu and Shenshu points.Compared with the control group,the stereotype score in the model group was significantly higher(respectively 63.77±5.67 scores;0±0 scores),with statistical significance(P<0.01),indicating that the model was successfully built.Compared with the model group,the stereotype scores of acupuncture group 1 and acupuncture group 2 were significantly reduced(respectively 33.32±5.17 scores;14.31±3.22 scores),indicating that acupuncture treatment is effective,and the effect of acupuncture group 2 is more obvious,with statistical significance(P<0.01).Results:From the evaluation of the effect of improving behavior,acupuncture at Dazhui and Shenzhu points is more effective than Pishu and Shenshu points.2.Detection results of monoamine neurotransmitters in serum and striatal receptor in brain tissueThe content of dopamine(DA)in serum of rats in model group was significantly higher than that in control group(respectively 821.63±86.10 ng/ml;416±91.21 ng/ml),with statistical significance(P<0.05),indicating that the model was successfully built.Tiapride group(594.13±11.97 ng/ml),acupuncture group 1(656.63±38.96 ng/ml)and acupuncture group 2(510.38±43.37 ng/ml)were significantly lower than the model group(P<0.05).Acupuncture group 2 was significantly lower than tiapride group.Compared with the control group,the expression of D1R and D2R in model group was enhanced(respectively 1.39±0.00 ?g/g;3.22±0.00 ?g/g),with statistical significance(p<0.05);Compared with the model group,the expression of D1R and D2R in acupuncture group 2 was weakened(respectively 0.93 ±0.00?g/g;0.92±0.01 ?g/g)?Results:Acupuncture group 2 had more obvious effect on regulating monoamine transmitter level and receptor expression.3.Detection results of amino acid neurotransmitters in serum and striatal receptor in brain tissueGlutamic acid(Glu)content in rat serum was not detected;The content of ?-aminobutyric acid(GABA)in serum of rats in model group was significantly lower than that in control group(respectively 156.00 ±1.64 ng/ml;172.18±1.58 ng/ml),with statistical significance(P<0.05),indicating that the model was successfully built.Acupuncture group 1 and acupuncture group 2 were higher than model group(respectively 159.92±1.21 ng/ml;166.15±1.92 ng/ml),of which the GABA content in serum of acupuncture group 2 increased more obviously,with statistical significance(P<0.05).Acupuncture group 2 was significantly higher than tiapride group(161.80±0.39 ng/ml),with statistical significance(P<0.05).Compared with the control group,the expression of NMDAR protein in the brain tissue of the model group was enhanced(respectively 3.06 ±0.02 ug/g;1.00 ± 0.00 ug/g),with statistical significance(p<0.05);Compared with the model group,the expression of NMDAR protein in acupuncture group 2 was decreased(1.67±0.03 ug/g),with statistical significance(P<0.05).Results:Acupuncture group 2 had more significant effects on increasing GABA content and decreasing NMDAR protein expression.4.Determination results of Serum Oxides and AntioxidantsIn this experiment,glutathione(GSH),superoxide dismutase(SOD)and malondialdehyde(MDA)kits were used to detect the corresponding indicators.The results showed that the content was negative and could not be statistically analyzed.Summary:Acupuncture for calming the wind and relieving spasm(needling Dazhui and Shenzhu points)has a good effect on improving twitch behavior of model rats,and its mechanism of action is related to the regulation of neurotransmitter imbalance and receptor expression.
