Pharmacodynamic Study Of Wenyun Mixture’s Therapy To Spleen Diarrhea Of Mice

Diarrhea is one of the pediatric common diseases. Spleen deficiency diarrhea generally has a long duration, which is a threat to the full body health and the normal growth and development of children. The famous old doctor of traditional Chinese medicine, Professor Wang Shouchuan proposes that characteristic of the pathogenesisis in pediatric spleen deficiency diarrhea is not only due to Spleen Qi deficiency, but also Spleen Yang deficiency. Its therapy should be adopted as warming Spleen Yang, thus he developed a prescription of Wenyun Mixture, which has a good effect on pediatric spleen deficiency diarrhea through the long-term clinical practice. However, there has been no laboratory evaluation and research on the prescription of Wenyun Mixture so far. This dissertation aims at characteristics of Wenyun Mixture. And this dissertation studied both Anti-diarrheal experiment and spleen deficiency diarrhea, and the latter is the study emphasis in mice.Objective:(1) Study on characteristics of Wenyun Mixture.(2) Study on the curative effect of Wenyun Mixture on Anti-diarrheal experiment of immature mice.(3) Study on the improvement of the general situation, protective effect on intestinal mucosal barrier and regulating effect on the intestinal flora.Methods:(1) Evaluate the characteristics and treatment of Wenyun Mixture through the basic theory, clinical efficacy and pharmacological studies in the way of literature and clinical summary with the tutor.(2) Investigate the curative effect of Wenyun Mixture on diarrhea of immature mice caused by sennae and castor oil respectively. Investigate the stool and inhibition on the small intestine of immature mice by carbon ink experiments.(3) Focused investigation on the curative effect of Wenyun Mixture on the spleen deficiency diarrhea of immature mice caused by rhubarb decoction. The model caused by rhubarb decoction was treated by Wenyun Mixture. Observation was contained with mental state, hair gloss, weight change, diarrhea index, et al.(4) Investigate intestinal pathology test by optical microscope and short chain fatty acid content by gas chromatography.Conclusion:(1) Characteristics of Wenyun Mixture consists of original prescription prominent Fang-yi, refined types. Wenyun Mixture has a significant effect on warming the spleen and treatment of children with spleen deficiency diarrhea.(2) Wenyun Mixture can be resistant to diarrhea of immature mice caused by folium sennae and castor oil respectively. Wenyun Mixture can improve the stool and inhibit rapid motility of the small intestine.(3) Wenyun Mixture plays a curative effect on mental state, hair gloss, weight change, diarrhea index of the spleen deficiency mice.(4) Wenyun Mixture plays a therapeutic role that can improve the intestinal mucosal injury and the balance of intestinal microflora.
