Research objective: Through the establishment of diabetic rats in right tibiafracture model, to observe the local injections of rhBMP-2and injection of rhBMP-2/PVP bone repair material impact on diabetes than fracture healing in rats, andcomparative research of liquid polyethylene pyrrole ketone solvents in diabetic rats infracture healing slow-release effect of BMP.Background: As the BMP-2degradated easily in bone engineering tissue, and thediabetic patients with lower rhBMP-2in the fracture healing process significantly,PVP(polyethlenepyrolindone) combined with rhBMP-2(bonemorphogenetic protein)is ideal for its slow-release effect in theory.Materials and methods:72healthy and2months old male SD rats,wererandomly divided into group A:18, only for non diabetic rats group (blank controlgroup), and more than54are induced after the destruction of pancreatic islet cells fordiabetic rats group (STZ), STZ group and random match in group B(18diabeticmodel group),group C(rhBMP-2groups of18) and group D(18rhBMP-2/PVPgroup).As four groups of diabetic rats were be right tibial fracture for diabetic ratstibia fracture model, rhBMP-2to effectiveness of gastrocnemius muscle note rhBMP-2solution (including rhBMP-2to5mg/Kg), rhBMP-2/PVP group to be in thegastrocnemius muscle note rhBMP-2/PVP composite slow-release solution(including rhBMP-2to5mg/Kg, PVP100mg/Kg), diabetic model group and blankcontrol group performed intraoperative gastrocnemius muscle physiological salineinjection capacity (such as solution).The fracture in2weeks,4weeks,6weeks were executed in each group,6rats right lower limb X-ray slice observation and fractureorganization HE staining, histological staining, ria method of determination of serumOC(ostepcalcin), alkaline phosphatase (alkaline osphatase, ALP), and then observecallus OB (osteoplast), and depicting the BMP-2release curve.Results: Fracture at the same time, X-ray examination results: each group partialshadow callus increased, which the amount of new bone scabs are more than A groupB, group C, group D and are less, most group D to produce callus, healing the ideal.Hematoxylin eosin staining were observed under light, according to the number ofOB significantly more than group B, group C, slightly lower than that of group A waslower than that in group D. Immunohistochemical staining observation: after Ki67index (%)4weeks after surgery to peak into decline, after each time point data show:peak above each group D. Biochemical measurements showed that OC concentrationresults show that the group D significantly higher than the other three groups,concentration of ALP results show that the D group had a significantly higher ALPcontent in each group. Bone repair the Ca content increases with the increase of time,content of statistical results show that the D> A> B> C. Histologic biopsy resultsobservations show that: the density of trabecular bone maturity and D group werehigher than the other three groups.Conclusion:1.Local injection of rhBMP-2/PVP slow-release composite canpromote healing in tibial fracture on diabetic rats.2. PVP is a relatively superiorinjectable sustained-release material liquid for rhBMP-2, and has a ideal slow-releaseeffect to rhBMP-2,and rhBMP-2/PVP slow-release composite on healing promotingeffect in fracture is better than that of local rhBMP-2function.
