Objective:To explore the expression of PCA3 gene in prostate hyperplasia and prostate cancer patients;compare the effect of urine PCA3 score in the diagnosis of prostate cancer in patients with different PSA levels to improve the accuracy of prostate biopsy and increase the early detection rate of prostate cancerMethods:Selected postoperative tissue specimens from patients who underwent prostatectomy and radical prostatectomy at the Second Affiliated Hospital of Kunming Medical University from May 2018 to November 2018.Prostate cancer tissue specimens were obtained through radical prostatectomy and prostate hyperplasia tissues Specimens were obtained by electrosurgical prostatectomy.All tissue specimens were pathologically diagnosed as prostate cancer or benign prostatic hyperplasia.The expression of PCA3 in prostate cancer and benign prostatic hyperplasia was measured and compared using RT-PCR.The patients who underwent prostate biopsy from November 2018 to December 2019 to the Second Affiliated Hospital of Kunming Medical University were selected,and the patients were divided into prostate hyperplasia group and prostate cancer group according to the results of prostate puncture.All the tested patients had their PC A3 detected and their PC A3 scores calculated from the initial urine after prostate biopsy.Count the age,FPSA,TPSA,and length of prostate diameter of the above patients before prostate biopsy,calculate FPSA/TPSA,prostate volume,compare the differences between the two groups of patients,and use the receiver operating curve(ROC)Analysis of the diagnostic value of urine PCA3 score in patients with different PSA levels for prostate cancer.Results:A total of 21 prostate tissue specimens were collected,including 11 cases of prostate cancer and 10 cases of benign prostatic hyperplasia.The results of RT-PCR showed that the expression of PC A3 in prostate cancer tissue was significantly higher than that of benign prostatic hyperplasia(P<0.01).A total of 113 patients with prostate puncture were included,including 49 in the prostate cancer group and 64 in the benign prostatic hyperplasia group.The differences in age,FPSA,and prostate volume between the two groups were not statistically significant(P>0.05);but the TPSA,FPSA/TPSA and PCA3 scores were statistically different(P<0.01).The receiver operating curve(ROC)analysis results showed that:in 113 patients with urine PCA3 score,the area under the curve for diagnosis of prostate cancer AUC=0.760,sensitivity 77.55%,specificity 68.75%(P<0.01);urine PCA3 score was PSA is located in patients with 4-1 0,10-20,20-50ng/ml,the area under the curve for diagnosis of prostate cancer AUC is 0.838,0.701,0.846(P<0.01);in PSA is located in 50-100ng/ml Among the patients,the area under the curve of PCA3 score for diagnosis of prostate cancer AUC=0.542(P>0.05).Conclusion:The PCA3 gene is highly expressed in prostate cancer tissues and is a good tumor marker for prostate cancer.Urinary PCA3 score has a good diagnostic effect on prostate cancer,and the diagnosis effect is the best among patients with PSA between 4-50ng/ml.
