Objective:By detecting the expression of KLF5in renal cancer tissues、adjacent tissues,toanalyze the expression level of KLF5in renal cancer tissues、adjacent tissues andexplore the relationship between the expression of KLF5in renal cancer and clinicalbiological characteristics.Methods:Source tissue:55cases of renal cancer tissues and10cases of adjacent tissues(above5cm away from the edge of the tumor specimens) were collected. Beforeoperation, all patients were not receiving radiotherapy and chemotherapy.1.Immunohistochemistry SP staining was used to detect the expression of KLF5,which contains55cases of renal cancer tissues and10cases of adjacent tissues.2. RT-PCR was used to compare KLF5mRNA expression in renal cancer tissuesto adjacent tissues of the same patients.Cases of renal cancer tissues which wereremoved by operation contrast with adjacent tissues whose distance was above5centimeter from the tumors. β-Actin used as the internal standard,was performed todetect the level of KLF5mRNA between renal cancer tissues and adjacent tissues.3. Western blotting was used to compare KLF5expression in renal cancer tissuesto adjacent tissues of the same patients. Cases of renal cancer tissues which wereremoved by operation contrast with adjacent tissues whose distance was above5centimeter from the tumors. β-Actin used as the internal standard,was performed todetect the level of KLF5mRNA between renal cancer tissues and adjacent tissues,image analysis.Results:1. Immunohistochemistry staining results showed that:(1)KLF5were expressed inrenal cell carcinoma and adjacent tissues.In renal cell carcinoma tissues, theexpression of KLF5was mostly diffuse,lost of polar expression,staining levels increased,and most expressed in the membrane and the cytoplasm; but,in adjacenttissues,the expression are only in the distal tubule、collecting duct cell membrane andcytoplasm, moderate staining intensity,and the climax was distributed. The positiverates of KLF5in renal cell carcinoma and adjacent tissues were significantly different(P<0.05),renal cell carcinoma was significantly higher than in adjacent tissue.(2)Thereis no significant difference in the relationship between the KLF5gene expression andthe gender,the age,tumor location as well as histological types (P>0.05), and nocorrelation with histological grades (P>0.05), there is a significant relationshipbetween the KLF5gene expression and clinical stage,and showing a positivecorrelation (P<0.05),the expression of KLF5in metastatic renal cancer tissues washigher than non-metastatic renal cancer tissues,the difference was statisticallysignificant (P<0.05).2. RT-PCR detection result showed that:The level of KLF5mRNA expression ishigher in renal cell carcinoma compared to adjacent tissues.The positive rate of KLF5mRNA expres-sion is100.0%but only30.0%in adjacent tissues.There is asignificant difference of KLF5mRNA expression between renal cell carcinoma andadjacent tissues (P<0.01).3.Western blotting detection result showed that: The result of the Image-pro plusVersion6.0analysis showed that the expression of KLF5protein in renal cellcarcinoma were significantly higher than in adjacent tissues (P<0.01).Conclusion:1.The overexpression of KLF5protein may play a role in initiation and/orprogression as well as the metastatic of renal cell carcinoma.2. The overexpression of KLF5protein in renal cell carcinoma was positivelycorrelated with clinical stage and metastasis of renal cell carcinoma.3. Detecting the expression of KLF5protein is important to assessing themetastasis and progression of renal cancer.
