| Objective:In the tumor cell,the key enzyme of glycolysis is highly expressed,and the tumor metabolism in lung cancer is also extremely active.KLF5 is a transcription factor involved in the development of a variety of tumors and is context-dependent.However,its role in tumor metabolism has not been clarified.This topic mainly discusses the changes in energy metabolism of A549 cells in anoxic environment.By observing the absorption of glucose,the production of lactic acid,the production of ATP and the changes of key enzymes of glycolysis,the relationship between KLF5 and key enzymes of glycolysis in A549 energy metabolism was investigated.This article aims to investigate the specific mechanism of glucose metabolism in lung cancer A549 cells and the effect of KLF5 on glucose metabolism in A549 cells in hypoxic environment.To further explore the potential targets of lung cancer treatment and the potential mechanism of action of KLF5 in lung cancer.Methods The A549 cell line was retained in the laboratory.A549 cells were cultured in 1640 medium containing 10% fetal bovine serum in 21% oxygen or 1% hypoxia.The ATP test kit was used to detect A549 under different environment.Intracellular ATP concentration,lactic acid test kit was used to detect the concentration of lactic acid in A549 cells of different concentration groups and control group.The glucose test kit was used to detect glucose uptake and intracellular glucose concentration in culture medium.The western blotting was used to detect the expression of glucose metabolism related proteins:LDHA,HK2,PKM2.Results 1.A549 cells can increase glucose uptake and concentration of lactate under hypoxic conditions.2.Hypoxia treatment can up-regulate the expression of key enzymes of glycolysis,HK2,PKM2 and LDHA in a concentration-dependent manner;the expression level of key enzymes is basically stable after 24 hours of treatment.3.Under normal-oxygen conditions,A549 oxidative phosphorylation and glycolysis exist simultaneously.Under hypoxia conditions,A549 mainly uses glycolysis as the main glucose metabolism.4.Knockdown or overexpression of KLF5 under normoxic conditions did not affect the expression of GLUT1,HK2,and PKM2 in A549 cells.5.Hypoxia can increase the expression of KLF5.6.Knockdown of KLF5 under 1% hypoxia can inhibit the absorption of glucose,the secretion of lactic acid and inhibit the production of ATP..7.HK2,PKM2,and LDHA was reduced by transient knockdown of KLF5 under hypoxic conditions.8.Overexpression of KLF5 under 1% hypoxic conditions can promote the absorption of glucose,the secretion of lactic acid and promote the production of ATP.9.Overexpression of KLF5 in 1% hypoxic conditions can up-regulate the expression of key enzymes of glycolysis,HK2,PKM2 and LDHA;Conclusions 1.Under hypoxic conditions,it can up-regulate the expression of key enzymes of glycolysis and promote the glycolysis of A549 cells to improve the survival rate of tumor cells.2.KLF5 can affect the expression of key enzymes in glycolysis of A549 cells and participate in the energy metabolism of tumor cells. |
