Epithelial ovarian cancer is the most common form of ovarian cancer and one of the most lethaly gynaecological cancers.The development and progression of ovarian cancer is a multi-stage process involving proliferation,apoptosis,angiogenesis,migration,invasion and metastasis.Therefore,understanding the molecular mechanism of ovarian cancer is of great significance for the prevention,diagnosis and treatment of ovarian cancer.In recent years,studies have found that a variety of lncRNA are involved in the development of ovarian cancer.The long non-coding RNA is a class of tissue-specific ncRNAs over 200 nt in length.lncRNA interacts with DNA,proteins and RNA to regulate gene expression at epigenetic,transcriptional and post-transcriptional levels.lncRNA plays an important role in many biological fields such as tumorigenesis,neuroscience and ontogeny,including proliferation,apoptosis,invasion,migration of tumer,resistance to chemotherapy.In this study,we screened the differentially expressed lncRNA: lncRNA PCM1 by sequencing the transcriptome of ovarian cancer cells.,the specific expression of lncRNA PCM1 in highly metastatic epithelial ovarian cancer was verified and highly expressed in tissues and serum of ovarian cancer patients.In vitro experiments such as CCK-8,scratch,flow cytometry and other in vitro experiments,we found that lncRNA PCM1 can promote the proliferation and migration of ovarian cancer cells and inhibits the apoptosis of ovarian cancer cells.we predicted the microRNA and microRNA target gene KLF5 interacting with lncRNA PCM1 through bioinformatics software,and further verified the relationship between the two luciferase reporter genes and Western blotting experiments.lncRNA PCM1 competitively binds to miR-153-3p,inhibits microRNA activity,and inhibits the degradation of target gene KLF5.lncRNA PCM1 contributes to ovarian tumorgenesis by directly regulating miR-153-3p/KLF5.These results provide a theoretical basis for the early diagnosis and treatment of ovarian cancer.
