| ObjectiveKrupple-like factor 5(KLF5)belongs to the KLF protein family,which contain three peptide modules of the c2h2 zinc finger type.As a basic transcriptional factor,KLF5 also play an essential roles in cell proliferation,differentiation,apoptosis and tumor progression which regulate gene expression through inhibiting or activating target gene expression.Many studies had indicated that high expression of KLF5 in CAFs is correlated with the progression and prognosis in many tumors.However,rare reports have sought to find whether increased KLF5 expression in CAFs in gastric cancer cells is involved in the cancer progression and its clinical significance.MethodsIn this study,we primary cultured the gastric cancer CAFs and gastric NFs.Immunohistochemistry was used to detect the KLF5 expression in GCAFs.The specific siRNA and KLF5-lenti were applied to explore the effects of the interaction between KLF5 expression of CAFs and gastric cancer cells on cell migration and invasion.In addition,120 samples of carcinoma tissue were selected randomly without any treatment before surgical resection from gastric carcinoma tissues.Collagenase digestion method was used to cultivate gastric cancer fibroblasts.Immumohistochemistry was applied to detect the expression of KLF5 in GCAFs from carcinoma tissues.The results were use for further analysis of correlation of a number of clinicopathological parameters including Age,Sex,Tumor size,Lauren’s classification,Grade,Invasivedepth,Lymph node metastasis and TNM staging as well as correlation with the factors.ResultsKLF5 was found that it is highly expressed in GCAFs,andthe over-expression of KLF5 in CAFS enhanced gastric cancer cell migration and invasion.To further indicate the clinical significance we found that the positive expression rate of KLF5 in GCAFs was 70%(84/120),KLF5 expression are significantly associated with invasive depth(P=0.016;P=0.01),lymph node metastasis(P=0.005;P=0.005)and TNM staging(P=0,016;P=0.001),but show no significant correlation with Age,Sex,Lauren’s classification,Grade.Colclusionour study indicate thatKLF5 is highly expressed in GCAFs and highly expression could significantly promote gastric cancer cell diligence and metastasis which is closely associated with poor prognosis. |
