Objective: To investigate the expressions of B7-H1And B7-H4in gallbladder carcinoma byimmunohistochemical SP method, and to analyze the relationship between the expression of them,and toinvestigate their regulating in the development of gallbladder carcinoma, it might provide new means ofprognosis evaluation, immunotherapy and gene therapy for gallbladder carcinoma.Methods:45cases from surgical specimens of patients with gallbladder carcinoma hospitalized in thegeneral surgery of the First affiliated hospital of Henan University from June in2002to June in2013, thosewho were diagnosed to be gallbladder carcinoma by pathology had not received preoperative chemotherapyor radiotherapy.20normal adjacent mucosa and20gallbladder adenoma served as normal control group,the expressions of B7-H1And B7-H4were detected by immunohistochemistry, the percentage of positivecell represented positive rank, the datas were analyzed by SPSS17.0, the statistical methods includedChi-square, Fisher’s exact test, McNemar test and Pearson contingency coefficient.Results:1.The positive rate of B7-H1in gallbladder carcinoma, gallbladder adenoma and normaladjacent mucosa was15.0%(3/20)、40.0%(8/20)、73.3%(33/45), respectively. B7-H1ingallbladder carcinoma was significantly higher than that in normal adjacent mucosa and gallbladderadenoma (P<0.05). However, there was no statistical significance between normal adjacent mucosa andgallbladder adenoma. In gallbladder carcinoma, the expression of B7-H1has no relation with some clinicalparameters including age, sex, tumor size, tumor location, gross morphology, pathological type and tumordifferentiation(P>0.05); But it had great relationships with the invasive depth(P<0.05), lymph nodemetastasis (P<0.05), TNM stages(P<0.05).2. The positive rate of B7-H4in gallbladder carcinoma, gallbladder adenoma and normal adjacentmucosa was54.5%(25/45)、15.0%(3/20)、0.0%(0/20); respectively, B7-H4in gallbladder carcinomawas significantly higher than that in normal adjacent mucosa and gallbladder adenoma(P<0.05). but therewas no statistical significance between normal adjacent mucosa and gallbladder adenoma. In gallbladdercarcinoma, the expression of B7-H4had great relationships with the invasive depth(P<0.05), lymph node metastasis(P<0.05), TNM stages(P<0.05) and tumor differentiation (P<0.05), but has no relations withsome clinical parameters including age, sex, tumor size, tumor location, gross morphology and pathologicaltype(P>0.05);3. In gallbladder carcinoma, B7-H1and B7-H4were expressed differently; but, the expression ofB7-H1and B7-H4was related (P<0.05).Conclusion:1.Both B7-H1and B7-H4were related with gallbladder carcinoma’s genesis.2.The high expression of Both B7-H1and B7-H4prognosticate the stronger ability of gallbladdercarcinoma to invasive and metastasize.3. B7-H1and B7-H4were co-expressed in gallbladder carcinoma, and detecting the expressions ofB7-H1and B7-H4will be good for diagnosis and judgement of prognosis. It might provide new means ofimmunotherapy and gene therapy for gallbladder carcinoma.
