Objective To investigate the expression of B7-H3and B7-H4and Their relationship in humancarcinoma of gallbladder.Methods The expression of B7-H3and B7-H4in the84samples was detected bystreptavidin-peroxidase immunohistochemical method, the expression of positive cells for B7-H3andB7-H4was evaluated, and their association with tumor classification, clinical grade, and recurrence wereassessed.Results In chronic cholecystitis tissue, B7-H3and B7-H4are not detected. In42cases ofcarcinoma of gallbladder,the positive rates of B7-H3and B7-H4expression were66.67%and69.05%respectively (P<0.05).B7-H3has increased expressions in low grade group(Well-differentiated-Differentiated)and highgrade group(Poorly-differentiated),58.06%and90.91%respectively (P<0.05).And The same was tosuperficial group(T1) and invasive group(T2-T4),33.33%and75.76%respectively (p<0.05).In primarygroup the positive Rate of B7-H3expression was53.57%,in recurrence group was92.86%(P<0.05).B7-H4has decreased expressions in low grade group(Well-differentiated-Differentiated)and high gradegroup(Poorly-differentiated))80.65%and36.36%respectively (P<0.05).And The same was to superficialgroup(T1) and invasive group(T2-T4),100%and60.61%respectively (p<0.05).In primary group thepositive Rate of B7-H4expression was85.19%,in recurrence group was40%(P<0.05).Expression of B7-H3was correlated with expression of B7-H4in carcinoma of gallbladder.Conclusions1.B7-H3was up-regulated in carcinoma of gallbladder and was closely related tothe patient’s pathological grade, clinical stage, tumor size, lymph node metastasis, liver infiltration and theoverall survival, regardless of the patient’s age and gender.The high expression of B7-H3may contribute tothe early diagnosis of carcinoma of gallbladder, and contribute to the assessment of postoperative survivaland recurrence.2. B7-H4were up-regulated in carcinoma of gallbladder and was closely related to the patient’spathological grade, clinical stage, regardless of the patient’s age, sex, tumor size, lymph node metastasis,liver infiltration and overall survival. B7-H4may plays an important role in the occurrence of gallbladder cancer.3.B7-H3and B7-H4may play a synergetic role in carcinoma of gallbladder. Combined tests areavailable for the diagnosis of carcinoma of gallbladder, assessment of the degree of malignancy andprognosis, and there may be a new target for carcinoma of gallbladder molecular targeted therapy.
