Background and Objective:Matrixmetalloproteinases9(MMP9) had realations with idiopathic pulmonaryfibrosis, chronic obstructive pulmonary disease (chronic obstructive pulmonarydisease, COPD), acute lung injury or acute respiratory distress syndrome and otherrelated diseases. MMP9was involved in the pathological process of coal workerspneumoconiosis. In addition, a significant increase of Matrixmetalloproteinases19(MMP19) was found in idiopathic pulmonary fibrosis. Pulmonary interstitial fibrosisis the basic pathological changes of pneumoconiosis, but the effection of MMP9andMMP19in the occurrence and development of pneumoconiosis is not clear. Thisstudy is to measure the peripheral serum levels of MMP9and MMP19in patientswith pneumoconiosis, and explore the association between MMP9and MMP19withpneumoconiosis, in order to provide clues and basis for screening biomarker of thesigns of the early pneumoconiosis.Material and Methods:1. Subjects: Ninety eight male patients with pneumoconiosis, including41cases of silicosis and57cases of coal worker pneumoconiosis, were enrolled assubjects. They were diagnosed by Xinxiang Institute of Occupational Medicine in2012~2013year. And the patients’ age were32~80years, and with the average age(57.29±10.83) years. Ninety eight healthy male physical examinees came fromHenan Institute of Occupational Medicine were used as controls, which aged32~80years with the average age (57.29±10.83) years.2. Serum collections: A fasting blood sample (3m1) was collected from theperipheral venous blood of each patient or control. Put the serum in the ordinarycentrifuge tube at room temperature for30min, and separated the serum stored in-80℃refrigerator.3. ELISA: The expression levels of MMP9and MMP19in serum from thesubjects were measured by enzyme-linked immunosorbent assay. 4. Internal quality control (IQC): Seven serum samples were choosed torepetition detect, and the seven samples were tested in every enzyme label plate. Inaccordance with the data to calculate the mean and standard deviation, and make aquality control chart.5. Pulmonary function tests: The Detection indexs were FVC, FEV1.0andFEV1.0/FVC%.Results:1. The serum levels of MMP9and MMP19in the patients with silicosis orcoal worker pneumoconiosis were all lower than that in the control group, and thedifferences were statistically significant (P<0.05). The serum MMP19levels of thepatients with silicosis were significantly higher than those of the patients with coalworker pneumoconiosis (P<0.05); there were no significant differences in serumMMP9expression between silicosis and coal worker pneumoconiosis (P>0.05).2. According to the research objects of different age, in the control group: theserum levels of MMP9in age between50and60years old group was significantlylower than the age less than50years old group, and the serum levels of MMP9inage more than50years old group was significantly lower than the age less than50years old group. The serum levels of MMP19in age between50and60years oldgroup was significantly lower than the age less than50years old group, and thedifference were statistically significant(P<0.05). According to the research objects ofdifferent age, in the patients with pneumoconiosis group: No obvious differenceswere observed in serum MMP9and MMP19levels among different age group(P>0.05).3. No obvious differences were observed in serum MMP9and MMP19levelsamong different stages of pneumoconiosis (P>0.05). No obvious differences wereobserved in serum MMP9levels among different group of exposure time (P>0.05),but the serum levels of MMP19in group of exposure for less than7years wassignificantly higher than the group of exposure20~28years or more than28years,and the difference were statistically significant (P<0.05).4. In the patients with pneumoconiosis group: No obvious differences wereobserved in serum MMP9and MMP19levels among different pulmonary function injury group (P>0.05).5. The serum MMP9and MMP19expression of the patients withpneumoconiosis were positively correlated with the control group (P<0.05); Theserum MMP9expression was negatively correlated with the stages ofpneumoconiosis (P<0.05); In the control group, the serum MMP9expression wasnegatively correlated with the age (P<0.05); In the patients with pneumoconiosisgroup: The serum MMP9expression was negatively correlated with the age(P<0.05), the serum MMP19expression was negatively correlated with the time ofexposure (P<0.05).Conclusion:MMP9and MMP19were involved in the occurrence and development ofpneumoconiosis. After further researches, the serum MMP9and MMP19levels maybe used as potential biomarkers for pneumoconiosis....
