Objectives To discuss the relationship between the polymorphism of metal matrix proteinases and the pneumoconiosis. We try to find the evidence of genetic sensitive markers of MMPs for screening the high-risk workers who was exposed to dust.Methods This study used grouped case-control study.We choosed 215 patients who have been diagnosed pneumoconiosis patients as a case group, and 241 workers exposed to dust who are nonpneumoconiosis as a control group. We used questionnaire to collected data. The venous blood DNA was used as the experimental material. The polymorphisms of MMPs were detected by the method of PCR-RFLP techniques. Use the χ2test to compare the diffrence between the two cases. The linkage disequilibrium coefficient of allele and haplotype frequencies were analysed with the SHEsis software.Results 1 In general The factors including age, cumulative dust exposure and smoking rate were all not significance(P>0.05). 2 The frequencies of MMP-1G-1607 G locus in case were 8.9%, 33.0% and 58.1%, compared with the control(9.1%、34.0% and 56.8%),the difference was not significant. The difference of the frequencies of genotype and allele in the silicosis and CWP were not significant between groups.(P>0.05). 3 The frequencies of MMP-2C-735 T locus C/C,C/T and T/T in case and control were 51.2 %,33.5%, 15.3% and 65.1%, 27.8%, 7.1%, the difference between the two groups was significant. The frequencie of allele were 32.1% for T in cases was higher than the frequencie in controls(21.0%). There were significant associations between the two groups(P<0.05). The difference of the frequencies of genotype and allele between silicosis and control was significant(P<0.05). The frequencies in CWP and control were44.1%, 36.3%, 19.6% and 61.1%, 28.6%, 10.3%, there was significant difference. The frequencies of allele T in CWP case was higher than the control, the difference between the the two groups was also significant(P<0.05). 4 The frequencies of MMP-3A-1171 A locus of genotype and allele in case of pneumoconiosis, silicosis and CWP and control had not significant diffrence(P>0.05). 5 The frequencies of MMP-7A-181 G locus A/A,A/G and G/G in case and control were 63.7%, 28.8%, 7.4% and 70.1%, 29.1%, 0.8%,there were significant difference. The frequencies of allele for G in case was higher than it in control.The significant associations were found between the two groups(P<0.05). The frequencies in silicosis and control were 74.3%, 18.6%, 7.1%, and 87.8%、12.2%' 0.0%, there were significant difference(P<0.05). The frequencies of allele in silicosis and control has significant difference. The frequencies of genotype and allele in CWP and control had no significant difference(P>0.05).6 The frequencies of MMP-12A-82 G locus A/A, A/G and G/G in case and control were 88.8%, 11.2%, 0.0% and 96.7%, 3.3%,0.0%, there were significant difference between the two groups. The frequencies of allele were 5.6% for G in case was higher than it in control(1.7%).The significant associations were found between the two groups. The difference of the frequencies of genotype and allele in the silicosis and CWP were significant between groups(P<0.05). 7 The phenomenon of linkage disequilibrium between 2G allele of MMP-1 and 6A allele of MMP-3 was not been found in this study population.The difference of the distribution of haplotypes between cases and controls was not significant(P<0.05).Conclusions 1 The study find that ther is association between gene polymorphisms of MMP-2C-735 T, MMP-7A-181 G and MMP-12A-82 G locus and the susceptibility of pneumoconiosis. The workers exposed to dust carrying the T/T genotype of MMP-2C-735 T locus, G/G genotype of MMP-7A-181 G locus and G/G genotype of MMP-12A-82 G locus will increase the risk of pneumoconiosis.2 There is no relationship between MMP-1G-1607 G and pneumoconiosis have been found in this study. The relationship between MMP-3A-1171 A genotypes and pneumoconiosis is also not been found too.3 This study find that the phenomenon of linkage disequilibrium is not exsisted between MMP-1 and MMP-3 gene. It is not been found that the four haplotypes were related to the pneumoconiosis.
