Background and Objective:Nasopharyngeal carcinoma (NPC) is a malignant tumor that comes from nasopharyngeal epithelium.It is happened over the world, especially in south of Chain. Radiotherapy is the most important method to cure the cancer, the more early the more well. However, NPC happens in a larval place, the symptom is not obviously in early. Searching for a new mark of nasopharyngeal to help diagnosis.NPC like others tumors, is polygene polystages, polysteps occurrence process with complicated mechanism.it relates numerous genes and proteins. The proto-oncogene c-myc is involved in cell proliferation, differentiation and apoptosis, and play an important role in tumorigenesis. Enolase-αencodes MBP-1, locating in nucleus. MBP-1 can bind promoter c-myc P2 to negative accommodate the expression of c-myc in nucleus.The binding can restrain tumorigenesis. BLU and MMP19 is Zinc domain-containing protein, lower expression in some tumers.In clinic, specimens were obtained by biopsy, usually the specimens are too small. According to above metioned consider,in this study,Tissue microarray and immunohistochemistry(IHC)were used to detect the expression of enolase-α,BLU and MMP19 in nasopharyngeal carcinoma (NPC). So that to evaluate the relationship between the expression level of enolase-α,BLU, MMP19 and tumorigenesis progress.Methods:We had collected 121 cases of NPC tissues,31 cases of adjacent nasopharyngeal epithelium and 20 cases of normal epithelium of nasopharyngeal which had been diagnosis by Dr. of the Department of Pathology of the Medical University of GungXi from 2003,2-2008,12,Tissue microarray and immunohistochemistry were used to detect protein expression of enolase-α,BLU and MMP19.The results were used to analyze the role of enolase-α,BLU and MMP19 among different clinic data (inluding age, gender, histology type, lymph node metastasis and clinical TNM stage) by stastistical methods.Results:(1)The normal positive expression of enolase-a in normal epithelium of nasopharyngeal, adjacent nasopharyngeal epithelium and NPC tissues were 94.4%,91.%7和52.2%, respectively. down-regulation of enolase-a in NPC was significantly lower than that of in normal epithelium of nasopharyngeal and adjacent nasopharyngeal epithelium (p<0.05).Expression of enolase-a protein detected in nuclei in NPC tissues was lower than that of in normal epithelium of nasopharyngeal and adjacent normal nasopharyngeal epithelium (p<0.05). In NPC cases, the normal expression of enolase-a was not correlated with age, gender, histology type, lymph node metastasis and clinical TNM stage (p>0.05) (2)The normal positive expression of BLU in normal epithelium of nasopharyngeal, adjacent nasopharyngeal epithelium and NPC tissues were 90%,100%和27%, respectively. down-regulation of BLU in NPC was significantly lower than that of in normal epithelium of nasopharyngeal and adjacent nasopharyngeal epithelium(p<0.05). In NPC cases, the normal expression of BLU was not correlated with gender, histology type and lymph node metastasis (p>0.05).The positive expression of BLU in age exceed 45 is lower than age less 45. The positive expression of BLU in TMN stagingⅠ+Ⅱcarcinnoma tissue is higher thanⅢ+Ⅳcarcinnoma tissue.The different has statistical significance (p<0.05).(3)The normal positive expression of MMP19 in normal epithelium of nasopharyngeal, adjacent nasopharyngeal epithelium and NPC tissues were 84.2%,84%和55.6%, respectively. down-regulation of MMP19 in NPC was significantly lower than that of in normal epithelium of nasopharyngeal and adjacent nasopharyngeal epithelium (p<0.05).In NPC cases, the normal expression of MMP19 was not correlated with age, gender, histology type, lymph node metastasis and clinical TNM stage (p>0.05)Conclution:The expression of enolase-α,BLU and MMP19 in NPC was lower than that of in normal epithelium of nasopharyngeal and adjacent nasopharyngeal epithelium. The lower expression maybe suggest that enolase-α,BLU and MMP19 had the close relation to carcinogenesis of NPC.Them wound be a new mark of NPC,helping to diagnosis and cure in NPC.
