Objective:To observe the clinical symptom signs, gastroscope signs and pathological changes of chronic atrophic gastritis (CAG) patients following Weiwei Granule and to assess the comprehensive therapeutic effects. To explore the effects of Weiwei granule on Bcl-2and PCNA expressions in gastric mucosa of chronic atrophic gastritis (CAG) rats and further investigate the mechanisms.Methods:The present research included clinical study and animal experimental study. In the clinical study,72cases of CAG patients which were accordance with criteria were randomized into treatment group and control group, with36cases in each. In the treatment group, Weiwei Granule were orally applied,20g each time, twice per day, half an hour before food intake; in the control group, Weifuchun tablets were orally applied,4tablets each time, three times per day, half an hour before food intake with warm water, each with3months as a therapeutic session. After one session, the results were re-checked by the same gastroscopy doctor and pathological doctor, and the clinical symptoms, gastroscope signs and pathological changes were compared and the comprehensive effects were assessed. In the animal experiments, in total fifty SD rats were randomized into normal group, modeling group, low-dose group (1.8g/d), middle-dose group (3.6g/d) and high-dose group (7.2g/d), with ten in each group. Besides normal group, rats in other groups were modeled as chronic atrophic gastritis with comprehensive method, and the treatments lasted for12weeks. With the immunohistochemistry method, the PCNA and Bcl-2expressions in the gastric mucosa were detected. Results:In the clinical study, the comprehensive effect in the treatment group was85.29%, which was higher than that of68.57%in the control group, with significant difference (P<0.01). After the treatments, the situations of gastroscope signs and pathological changes in the treatment group were better than those in the control group (P<0.01). However, there were no significant difference of the clinical symptom improvement between the two groups (P>0.05). In the animal experiments, compared with the modeling group, the0D value of Bcl-2in all the treatment groups of Weiwei granule were obviously decreased (P<0.01). Compared with the low-dose group, the0D value of Bcl-2in middle-dose group were obviously decreased (P<0.01). Compared with the middle-dose group, the OD value of Bcl-2in high-dose group group were obviously decreased (P<0.01). Compared with the modeling group, the PCNA expression in all the treatment groups of Weiwei granule were obviously decreased (P<0.01). Compared with the low-dose group, the expression of Bcl-2in middle-dose group were obviously decreased (P<0.01). Compared with the middle-dose group, the expression of Bcl-2in high-dose group were obviously decreased (P<0.01).Conclusion:For the CAG patients, Weiwei Granule could improve the clinical symptoms, gastroscope signs and pathological situations, which has definite therapeutic effects. Weiwei granule might inhibits the Bcl-2and PCNA expressions in the gastric mucosa of CAG rats and regulates the cellular proliferation and apoptosis hence improves the abnormal situation of the gastric mucosa and prevents the CAG further transformation into gastric carcinoma. Hence, Weiwei Granule is applicable and feasible in the treatment of CAG, which is worthy of being further popularized in clinic.
