Objective:To explore the mechanism of Xiefuning Granule on chronic non-atrophic gastritis by studying the expression of Janus activated kinase 2?JAK2?,P-Janus activated kinase 2?P-JAK2?,signal transducer and activator of transcription 3?STAT3?and interleukin-6?IL-6?and cell apoptosis in rats with chronic non-atrophic gastritis.To improve the etiology and pathogenesis of CNAG,and provide experimental basis for clinical application of Xiefuning Granule on CNAG and development of new drugs.Methods:Fifty SPF SD rats were randomly divided into normal group?CONT?,model group?CNAG?,high dose group?HIG?,low dose group?LOW?and omeprazole group?OME?.In the normal control group,the rats were given ordinary feed.The other four groups were fed with different concentrations of ammonia water and starvation disorders to establish CNAG animal models.In the Xiefuning group,0.5g.mL-1Xiefuning solution was given 2mL,1mL,and in the Omeprazole group,0.67mg.mL-1Omeprazole solution was given 2mL.In the model group,the rats were perfused with the saline 2mL,once a day,for 15 days.Serum and gastric tissue of rats were taken after administration.To measure the expression of JAK2,P-JAK2 and STAT3 in rat gastric tissue by immunohistochemical staining;To measure the levels of JAK2,P-JAK2,STAT3 and IL-6 in rat serum by enzyme-linked immunosorbent assay?ELISA?;To measure the expressions of JAK2,P-JAK2,STAT3 and IL-6 in rat gastric tissue by Western blot;To analysis the apoptosis of rat gastric tissue by TUNEL.Results:?1?In the CNAG group,the gastric mucosa had complete margin,normal cell and gland structure,a small amount of lymphocyte was scattered,and no inflammatory cell infiltration was observed.In the CNAG group,gastric mucosa margins were irregular,epithelial cells necrosis,a large number of inflammatory cells infiltration in the lamina propria of mucosa,and capillary dilatation and congestion.The number of inflammatory cells in the lamina propria of gastric mucosa decreased and the arrangement of gastric glands tended to be regular in the drug groups.?2?The expressions of JAK2,P-JAK2,STAT3 and IL-6 in serum and gastric tissue of the CNAG group rats were significantly higher than those of the CONT group?P<0.05?.The expression of the above proteins in the drug groups was significantly lower than that in the CNAG group.The expression of histone protein in the HIG group and the OME group was significantly down-regulated,while that in the LOW group was less down-regulated?P<0.05?.?3?The apoptotic rate of gastric tissue in the CNAG group was significantly higher than that in the CONT group.The apoptotic rate decreased in the drug groups,especially in the HIG group?P<0.05?.Conclusions:Xiefuning Granule can inhibit the opening of JAK2/STAT3Signaling Pathway,down-regulate the expression of JAK2,P-JAK2,STAT3 and IL-6 in serum and gastric tissue of rats with CNAG,inhibit the apoptosis and inflammatory reaction of gastric tissue of CNAG rats,and play a role in the treatment of CNAG.
