Genetic Polymorphism Study Of Y-SNP In Yunnan Bai Male Population

[Objective] This study is based on phylogenetic tree of the Y chromosome, choosing43Y-SNP loci in O, C, N, using MassARRAY technology to research Y-SNP loci, acquiring the haplogroup frequency of Yunnan Dali Bai male population and the structure of the genetic condition, to establish Y-SNP-based DNA database and to lay the foundation for forensic applications.[Methods] Under the principle of informing voluntary and requiring their consent, the access standards isconduct strictly （male individuals, within three generations are Bai, unrelated）,5ml peripheral blood was collected from278cases of Yunnan Bai individuals in Yunnan Dali and the surrounding counties.After adding EDTA-Na2for anticoagulant, the MassARRAY assay was used to test43SNP loci in Y chromosome and statistical analysis were performed on SPSS19.0.The paper was used MassARRAY technology, stepwise screeninged samples by analysing O branch P101, L682, M175, M324, M134,2611, M50（MHO）, P164, Page23（M117）, M164, M162₁, Page20（M119）, P201, F706, M312, P31, Pk4, F238, M95, L127.1, P49, M7, F11, M88, M159, M188（M113）, M122, M101, P203.1loci,analysing C branch V20, M347, M38, M105, M216, M356, M210loci, and analysing the N branch L666, M231, L729, P189.2, L708, L732, M46（P105） sites, a total of43SNPs were in screening analysis.[Results] Obtain Y-SNP haplotypes and haplotype frequencies in278unrelated Bai males.28Y-SNP haplotypes were detected.the Y-SNP haplotype diversity of Yunnan Bai male population was0.0642、M175、M324、M134、P164、P201in the population haplogroup distribution frequency is higher. The haplogroups was not detected such as P101、L682、M50、M164、M1621、M312、P49、V20、M347、M38、M105、P189.2、 L732、M356、M210. For43Y-SNPs loci, the genetic diversity ranged from0.0000to0.6753.[Conclusions] Based on the human phylogenetic tree of the Y chromosome, the43Y-SNPs loci of278cases Yunnan Bai male sample was analysed.28Y-SNP haplogroups,haplogroup frequency distribution and genetic structure information was obtained.The result was the same with other minorities haplogroup distribution frequency analysis which had been reported. Not only can help to understand the origin of Bai and the relationship between Bai with other minority,help to study structural differences among ethnic genome and the genetic significance of the difference, help to provid evidence for human evolution and biology of all ethnic origins, migration and dividing in China, and try to establish with the MassARRAY analysis and detection methods to provide relevant data to Yunnan Y-SNP studies, while provid data for the establishment of the Yunnan-based DNA databases.
