Objective:Explore the relationship of SNPs of PCSK9 p.Arg93Cys(rs151193009)?LDLR p.Arg257Trp(rs200990725)?CETP p.Asp459Gly(rs2303790)?PKD1L3 p.Thr429Ser(rs7185272)? PNPLA3 p.Ile148Met(rs738409)?TEAD2 p.Asp12Asn(rs142665148)with gallstone disease,and whether the differences of rates of locis exist between the groups.Method: We collect 5 ml venous blood and gallstones from 100 patients which are treated as gallstone group and collect 5 ml venous blood from 30 normal people which are treated as control group.Use Fourier transform infrared spectrum to detect the components of gallstones to classify the gallstone group into two groups,the Cholesterol-type gallstone group and the Non cholesterol-type gallstone group,then use MassARRAY mass spectrometer to detect the mutants of locis in three groups' blood and analysis whether the differences exist between the groups.Result:1.There is significant difference between gallstone group and control group in the rate of mutant of TEAD2(rs142665148(P=0.039<0.05).The rate of mutant of TEAD2(rs142665148)in gallstone group is significant higher than that in control group;There are no significant differences between gallstone group and control group in the rate of mutant of PKD1L3(rs7185272)?PNPLA3(rs738409)?PCSK9(rs151193009)?LDLR(rs200990725)?CETP(rs2303790)(P>0.05).2.The rate of mutant of,PKD1L3(rs7185272),PNPLA3(rs738409)in Non cholesterol-type gallstone group are significantly higher than that in Cholesterol-type gallstone group.3.There are no significant differences between Cholesterol-type gallstones group and Non cholesterol-type gallstones group in the rates of mutant of PCSK9(rs151193009)?LDLR(rs200990725)?CETP(rs2303790)?TEAD2(rs142665148)(P>0.05).Conclusion:1.SNPs of TEAD2(rs142665148)?PKD1L3(rs7185272)?PNPLA3(rs738409)have association with gallstone diseases.2.SNPs of PNPLA3(rs738409),PKD1L3(rs7185272)have association with Non cholesterol-type gallstone diseases.SNPs of TEAD2(rs142665148)have associations with Cholesterol-type gallstone diseases and Non cholesterol-type gallstone diseases.
