Objective:Normal human Y chromosome is contemplated by the None recombinating Y chromosome (NRY95%)and PAR (5%),Male meiosis, PAR with X chromosome recombination.NRY was hemizygous haploid paternal inheritance,Unless a mutation.Otherwise NRY genetic markers from parents to offspring is Constant,thus mutated offspring will be cumulative.Therefore,the Y chromosome plays an important role in the study of human origin and evolution. Y-SNPs of genetic markers are two alleles of the genetic markers and the number of SNPs is huge,and the frequency is very dense distribution,Locus mutation rate per generation in about10-8,Almost no revertant,therefore, it plays an important role in the study of human origins and evolution.On this basis, we can draw the evolutionary tree of human,which can react evolution and migration of human,the study of the Y chromosome support the modern East Asian population from African origins,the results supported East Asia crowd is mainly by differentiated from O.In this study, by studing the evolutionary tree of the Y chromosome system,Screening out the specific sites in Yunnan Han population,then using this feature,locked the suspect in which the crowd,to the detection of actual cases.Methods:Three milliliter venous blood sample is obtained from each male patient. After adding EDTA to prevent the blood from coagulating, DNA was extracted, genotype and allele frequencies of polymorphism in SNPs of Y chromosome evolution tree were examined by SNaPShot and direct sequencing analysis of DNA technique in500male patients. All statistics were performed on Direct counting method.Results:416cases of O branch samples,28cases of C branch samples,34cases of N-branched samples a total of478samples:M117、002611、M159、M113、P164、M7、 M134、M119、P31、M95、M122、M88、M324、P201、M121、M164、M110、P203、 P101、M105、P103、M38、M217、M347、M356、P55、LLY22g、M128、P43、 P105、M178were detected.Each locus has mutation samples:M117:104samples;002611:39; M159:4; M113:2; P164:12; M7:6; M134:48; M119:12; P31:38; M95:17; M122:8; M88:9; M324:3; P201:9; M121:4; M164:0; M110:20; P203:21; P101:35; P103:17.LLY22g:0; M128:5; P43:0; P105:25; M178:5;M105:1; M38:0; M217:0; M347:26; M356:1; P55:0.Counting statistics for all samples,the entire Yunnan geographical distribution Y-SNP haplogroups frequency of O3a3c1-M117、03a4*-002611、O3a3a-M159、O3a3bl-M113、 O3a3b2-P164、03a3b-M7、3a3c-M134、O1a*-M119、O2*-P31、O2a*-M95、O3*-M122、 O2a1-M88、O1a2-M110、O3a*-M324、O3a3*-P201、O3a1-M164、O3a2-M121、 O3a4a-P103、O1a1-P203、O3a3c2-P101、N1*-LLY22g、N1b-P43、N1a-M128.
