Objective To evaluate the association of the polymorphism of theHLA-Ⅱgene and multiple sclerosis (MS) in Chinese population.Methods A computer-based online search was conducted inMedline(1950~2013),Embase(1980~2013),TheCochraneLibrary,CBMdise(1978~2013),CNKI(1970~2013),WanFang (1979~2013).Manual searching relevant journalssuch as Journal of Clinical Neurology, Chinese Journal of Neurology, Chinese Journalof Neuroimmunology and Neurology, Journal of Apoplexy and Nervous Diseases,Chinese Journal of Clinical Neurosciences etc to collect all world case-control studiesaddressing of the association of the polymorphism of the HLA-Ⅱ gene and multiplesclerosis in Chinese population. The quality of included studies was assessedaccording to the criteria recommended by the Cochrane Handbook for SystematicReviews of Interventions and data were extracted by two reviewers independently.We made meta-analysis, quantitative qualitative comprehensive analysis, funnel plotanalysis for the study results. In the meta-analysis, fixed or random potency modelwas chosen according to the heterogeneity. Meta analysis was conducted by TheCochrane Collaboration’s RevMan5.2software. Estimated the evaluation index by95%confidence interval and made α=0.05for the size of test.Results Fifteen case-control studies were included after screening andapplication of inclusion and exclusion criteria,including599MS patients and1127controls.META analysis showed:(1)Compared with control group, MS patientHLA-DPB1*1602gene expression is rather high (OR=2.35,95%CI (1.43,3.86),P=0.0007);(2)Compared with control group, the HLA-DQB1*0601, DRB1*0405, DRB1*1501,DPB1*1301of MS gene expression shows no value to statistics. Conclusion The limited evidence at present show that HLA-DRB1*1602gene liability,but the relationship between HLAⅡ gene and MS is still couldn’t beconfirmed.It should be further confirmed through more samples,more rigorouslydesigned and high quality inveseigation.
