【Objective】Research on cancer associated protein B cell lymphoma-2(Bcl-2),ubiquitinprotein ligase MDM2and cyclin-dependent kinase inhibitor P21in normal humanoral mucosa,oral leukoplakia and oral squamous cell carcinoma of human,expr-ession in squmous cell carcinoma further in-depth study of the development ofmechanisms to provide a scientific basis for oral squamous cell carcinoma.【Method】Apply two-step method of immunohistochemistry to detect the expression of Bcl-2,MDM2and P21in15cases of normal oral mucosa,25cases of oral leukoplakia and25csaes of oral squamous cell carcinoma. Immunohistochemisty and quantitative picture of the software relative quantitative analysis anstatist-ical and software are applied to analyze the experimental results.【Result】1.The expression of Bcl-2and MDM2in normal oral mousa,simple hyperplasia leukoplakia, hyperplasia leukoplakia and squamous cell carcinoma of human is gradually increasing.(P﹤0.05).2.The P21protein in normal oral mousa and simple hyperplasia leukoplakia most positive expression,and lower expression in hyperplasia leukoplakia and sq-uamous cell carcinoma. The statistical analysis showed that expression differe-nces between normal oral mucosa and simple hyperplasia leukoplakia of P21protein was not statistically significant. The normal oral leukoplakia,oral squam-ous cell carcinoma differential expression of P21protein was statistically signi-ficant.3.The correlation of Bcl-2,MDM2and P21:In hyperplasia leukoplakia and sq-uamous cell carcinoma, a postive relation was found between Bcl-2and MDM2expression, and no relation was found between Bcl-2and P21expression or P21and MDM2expression.【Conclusion】1. The expression of Bcl-2and MDM2in normal oral mousa,simple hyperplasia leukoplakia, hyperplasia leukoplakia and squamous cell carcinoma of human is gradually increasing. While the P21protein in normal oral mousa and simple hyperplasia leukoplakia most positive expression,and lower expression in hyperplasia leukoplakia and squamous cell carcinoma.These findings indicate that Bcl-2、MDM2and P21expression are correlated with the oral squamous cell carcinoma of human. Bcl-2、MDM2and P21protein can be used as the maker to predict behavior of squamous cell carcinoma2.In oral dysplastic leukoplakia and oral squamous cell carcinoma tissues, Bcl-2protein and MDM2protein expression was significantly positively correla-ted,suggesting that the two may have a synergistic effect in the carcinogenesis of oral mucosa.3.Although Bcl-2、MDM2and P21protein are associated genes with P53gene,but by statistical analysis shows that no relation between Bcl-2and P21expression or P21and MDM2expression,suggesting that P21protein on carcin-ogenesis mechanism of action is different from both.
