Objective: To detect human papilloma virus(HPV)genotypes and the histological expression of p16 protein in oral leukoplakia(OLK)and oral squamous cell carcinoma(OSCC).To explore the role and correlation of HPV and p16 in the development of OLK and OSCC.Methods:(1)Polymerase chain reaction(PCR)and diversion hybridization technology were utilized to detect 37 kinds of HPV genotypes in oral exfoliated cells from 30 cases of the normal mucosa,103 cases of OLK and 30 cases of OSCC,respectively.Clinicopathological characteristics of all subjects were also analyzed.(2)Expression of p16 was assessed by immunohistochemical analysis in the paraffin-embedded tissue from these OLK and OSCC.15 control cases of normal mucosa were excised from the impacted teeth extraction or dental implants surgery.The differences of p16 among these groups and the relationship with HPV were analyzed by the SPSS.Results: In normal mucosa,HPV detection rate was 3.33%(1/30)and the positive HPV was HPV68;In OLK,the rate was 4.85%(5/103),4 cases were detected with 1kind of HPV-positive,respectively HPV18?39?40 and 82,1 case was detected with 3kinds which were HPV35,44,51;In OSCC,the rate was 3.33%(1/30),and the positive was HPV67.There was no statistically significant difference among the groups(P> 0.05).From the normal,OLK to OSCC,p16 expression level gradually decreased,and the difference between the groups was statistically significant(P<0.05).There were 43 cases of p16 overexpression in OLK,of which 5 were HPV-positive.Only 1 in OSCC was p16 overexpression,while HPV was positive.Conclusion:(1)With the malignancy of oral epithelial tissue,p16 expression gradually decreased.(2)A poor concordance between p16 and HPV suggests that p16 expression should not be independently used as a marker for HPV infection in OLKand OSCC.(3)A low prevalence of HPV was detected in OLK and OSCC though different types of HPV can be detected.Whether HPV infection is involved in OLK and OSCC remains to be further studied...
