Objects: This study investigated the possible effective parts of Tibetan Oxytropisfalcata Bunge on improving myocardial ischemic-reperfusion injury in rats.Methods: We extracted the parts of ethanol extracts, chloroform and ethyl acetate ofthe Tibetan O. falcata Bunge by the way of first dissolving, second heating, thirdfiltering and so on. SD rats were randomLy divided into several groups: the shamoperation group, the IR model group, the control group, the ethanol extraction highdose group, the ethanol extraction middle dose group, the ethanol extraction low dosegroup, the chloroform high does group, the chloroform low dose group, the ethyl highdoes group, and the ethyl low dose group which accounted to ten groups. Aftercontinuously fed ten days by gavage once a day at9in the morning, the way we builtthe rats’ model construction of MIRI was via ligating rats’ the left anterior descendingwhich is in the coronary artery of hearts. When it went40minutes by reperfusion, wewould determine the model’s contents of LDH, CK, SOD, GSH-Px, and MDA in itsserum; content of NO and activity of NOS, iNOS in its myocardial tissues; and theultrastructural changes of MIRI rats should also be observed.Results:⑴Compared with the model group, the ethanol extracts high, middle andlow dose, chloroform high and low dose and ethyl high and low dose can reducesignificantly the elevated level of ST in ECG which caused by MIRI in rats(P<0.01);reduce the release of LDH and CK in the serum(P<0.05);enhance theactivity of SOD and GSH-Px in myocardial tissue and decrease the content of MDA(P<0.05,P<0.01);significantly depress the activitity of NOS, iNOS (P<0.05,P<0.01)and decrease the level of NO(P<0.05,P<0.01).⑵The level of its effectsis proportional to the doses of gavage.⑶Compared with the ethanol extraction highdose, the chloroform high and low dose and ethyl high and low dose have statisticalmeaning on reducing the ST level (P<0.05); the chloroform high and low dose reduce the release of LDH and CK have statistical significance (P<0.05), and the ethyl highand low dose have the statistical meaning of reducing LDH(P<0.05), significantstatistical meaning of reducing CK(P<0.01); the chloroform high dose and the ethylhigh dose have the statistical meaning on enhancing activity of SOD and GSH-Px andthe reducing contents of MDA(P<0.05); the chloroform high dose and the ethyl highdose have meaning on depressing the activitity of NOS, iNOS (P<0.05)anddecreasing the level of NO(P<0.05).Conclusion: The research had shown that, the main active ingredient of thechloroform and ethyl acetate of O. falcata Bunge is flavonoids[7,21-22], and theexperiment demonstrates that the effects of the chloroform and ethyl acetate of O.falcata Bunge is better than the ethanol extracts in impriving the myocardialischemia-reperfusion injury. Therefore, we believe that flavonoids could be theeffective parts of O. falcata Bunge against myocardial ischemic-reperfusion injury.However, the exact active ingredients and mechanism still need to have furtherresearches to prove.
