Objective: To study the protective effects of Tibetan medicine Oxytropis falcata Bunge(OFB)extractive on hypoxia/ reoxygen(H/R)injury in H9_C2 cardiomyocytes.Methods: Three gase incubator culture method was adopted to establish H9_C2 cardiomyocytes hypoxia/ reoxygen injury model.The experiment was divided into nine groups: normal control group(C);H/R control group(H/R);H/R control group plus seven drugs(H/R+OFB);according to Oxytropis falcata Bunge extract with the final concentration of 0.1,1,10,100,300,600,1 000 mg/L at the start of hypoxia/reoxygen.(1)Colorimetric assay was used to detect cell viability for cell injury,using the MTs method for determining the influence of drugs on cell vitality.Detecting the influence of Oxytropis falcata Bunge ethyl acetate methanol water column pressure components and chloroform extract components with different dosage on H9_C2 myocardial hypoxia/reoxygen injury.(2)It was found that the concentration of 300 mg/L was the best for drug The expression of Caspase-3 apoptosis protein by Western blotting method.Results:(1)Compared with the group H/R,H/R+OFB group significantly increased hypoxia/ reoxygen H9_C2 cardiomyocytes viability(p<0.05);(2)Caspase-3 apoptosis protein expression was no significant difference in H/R+OFB group compared with the H/R control group(p> 0.05).Conclusion:(1)Tibetan medicine Oxytropis falcata Bunge by increasing hypoxia/reoxygen injury of H9_C2 cardiomyocytes play a protective role.(2)Oxytropis falcata Bunge has protective effects on hypoxia / reoxygen injury of H9_C2 cardiomyocytes,but the mechanism is not through Caspase-3 pathway,but not with Caspase-3.
