Objective: This study investigated the information of in vitro and in vivo compounds,which were from different parts of Tibetan Oxytropis falcata Bunge,and its possible changes in rats.Methods: We extracted the parts of ethanol extracts,ethyl acetate,chloroform,water and N-butanol of the Tibetan O.falcata Bunge by the way of first dissolving,second heating,third filtering and so on.The chemical constituents which were from different extractions of Tibetan O.falcata Bunge were analyzed to determine the information of the compounds by LC-MS.These rats which were fed for 7 days were divided into four groups,its were the ethyl acetate extraction(EAEO)group,the chloroform extraction(CHEO)group,the N-butanol extraction(ZDCO)group,and the water extraction(EDX-S)group.There were 6 rats in every group,male and female each half.After drug administration,the 0.5 mL blood was collected from the vena ophthalmica according to the following times 0,30,60,90,120,180,240,360,480,720,1440 min,respectively.The chemical constituents were analyzed to determine the information of the compounds by LC-MS,and the possible changes in vivo of rats.The liver and intestinal fecal samples were collected after the last blood collection,and we studied the liver microsome metabolic process and enteral transformation experiment.The chemical constituents were analyzed to determine the information of the compounds by LC-MS,and the possible changes in vivo of rats,respectively.Results:(1)By the detection of LC-MS,This experiment found that: There are 15 chemical compositions in EAEO,including 12 flavonoids,1 glycoside compound,2 other compounds;There are 18 chemical compositions in CHEO,including 13 flavonoids,1 amide compound,1 glycoside compound,3 other compounds;There are 40 chemical compositions in EDX-S,including 24 flavonoids,2 amide compound,1 glycoside compound,13 other compounds;There are 14 chemical compositions in ZDCO,including 10 flavonoids,1 amide compound,3 other compounds.(2)Some flavonoids which are from different extractions of Tibetan O.falcata Bunge,including Genistein,Chrysanthemum,Citrinin,Quercetin,Ratin,7-hydroxyflavone,7-methoxyflavone,2'-hydroxy-4'-methoxychalcone,7-hydroxy dihydroflavone,2',4'-dihydroxy chalcone,may be involved in a series of biochemical reactions in rat plasma and may be involved in multiple phase I responses,which includes hydroxylation,methylation and so on.(3)Some flavonoids which are from different extractions of Tibetan O.falcata Bunge,including Myricetin,3',4',5,7-tetrahydroxybisflavone and 2',4'-dihydroxy chalcone,may be involved in a series of liver metabolic reactions and may be involved in multiple phase I responses,which includes hydroxylation,methylation and so on.(4)Some flavonoids which are from different extractions of Tibetan O.falcata Bunge,including Myricetin,Quercetin,7-methoxyflavone and 7-hydroxy dihydroflavone,may be involved in a series of Intestinal metabolic processes and may be involved in multiple phase I responses,which includes hydroxylation,methylation and so on.Conclusion: The compounds,which are from different extractions of Tibetan O.falcata Bunge and may be involved in the metabolic process in vivo of rats,are mainly flavonoids.Therefore,we consider that the total flavonoids may be the main active ingredients,which have its pharmacological and pharmacological effects in vivo.The main metabolism pathway for flavonoids involved in in vivo may be the phase I reaction,which includes hydroxylation,methylation and so on.This study confirm that flavonoids include Myricetin,Chrysanthemum,Citrinin,Quercetin,Ratin,Genistein,7-hydroxyflavone,7-methoxyflavone,2'-hydroxy-4'-methoxychalcone,7-hydroxy dihydroflavone,3',4',5,7-tetrahydroxybisflavone and 2',4'-dihydroxy chalcone,which may be involved in metabolizing in vivo.
