The Effect Of Yiqi Mixture On Expression Of Serum Complement C₃, IgM, IFN-γ Of Spleen-deficiency In Rats

Objective：According to the TCM theory, using the composite method of rhubarb andrestricted diet to establish rat models of spleen-deficiency, by observing the effect ofYiqi Mixture on the levels of serum IgM, complement C3, IFN-γ level, and thepathological changes of the spleen tissue after drug intervention, Si Junzi Decoctionas control, to investigate the immunomodulatory effect and mechanism of YiqiMixture.Methods：Fifty SD rats of clean grade,6W age, half male and half female, wererandomly selected16as control group, half male and half female, the rest of rats wereincluded in the model of spleen-deficiency group. Spleen-deficiency model wasestablished by rhubarb and restricted diet composite method, a total of21days. Afterthe modeling, the model was successfully established in rats were randomly dividedinto model group, Yiqi mixture group, the Si Junzi Decoction group,8rats in eachgroup, half male and half female, according to the experimental design for medicine,a total of21d. With drug intervention of21days,detect the serum complement C3,IgM, IFN-γ, and paraffin sections of spleen tissue.Results：1.2-3days later, the model of spleen-deficiency rats, loose stools.1w later,diarrhea or loose stools, curled up, squinting, stooping, diet and activity weresignificantly reduced; after3W modeling, that above obvious change. After modelestablishment, compared with the control group, the rats in the model group reducedweight, the swimming time shorten, spleen wet weight and spleen wet weight indexdecreased, there were significant differences (P<0.01), the model establishedsuccessfully.2.Compared with the control group, spleen-deficiency model rats serumIgM, C3content is blank group increased significantly (P<0.01), serum levels ofIFN-γ was significantly decreased than those of blank group (P<0.01). With thetreatment of3W, compared with model group, the content of the treatment ratsserum IgM, C3decreased significantly (P<0.01), but the serum levels of IFN-γ rosesignificantly (P<0.01); the amount of treated groups,the effect have no significant difference (P>0.05).3.Pathological of spleen tissue shows: compared with the modelgroup, the treatment groups of spleen tissue of lymphocytes reduce, lymphaticsheath, central arterial structure returned to normal.Conclusion:1. Yiqi mixture can reduce C3content of serum complement IgM, rat,elevated serum levels of IFN-γ.2.Yiqi mixture can improve pathological injury ofspleen-deficiency syndrome and spleen tissues of rats.3.Yiqi mixture can obviouslyrestore immune function of spleen-deficiency rats, may be associated with increasedserum levels of IFN-γ.4.This study deepens the understanding on the relationbetween the spleen-deficiency and immunity, the clinical and experimental studieson the effective use of dialectical argumentation in nature medicine, help to the studyof TCM theory deeply, the research and development of Chinese medicine andclinical significance.
