Objective:According to Traditional Chinese Medicine (TCM), spleen qi deficiency (SQD) is purported to be the main pathogenesis of allergic rhinitis (AR), establish on spleen-deficiency disease and syndrome compound model of allergic rhinitis, to observe on spleen-deficiency condition in the morbidity of allergic rhinitis'characteristics and the nasal mucosa's pathological changes. Using traditional Chinese medicine of tonifying the spleen and the qi and desensitization to drug intervention, which from pathology, inflammation angle to evaluate the effect of Yiqi Tuoming Prescription on spleen-deficiency allergic rhinitis and function mechanism. Providing the basis for clinical use of yi-qi desensitization method to treat allergic rhinitis.Methods:Normal SD rats were randomly divided into normal controls, allergic rhinitis (AR) group, spleen qi deficiency (SQD) model group, spleen qi deficiency combined with allergic rhinitis (AR+SQD) and AR+SQD treated with TCM. The AR group was modeled by using ovalbumin as the sensibiligen. The SQD group was underwent rhubarb purgation, eating and drinking without temperance and overstrains. The AR+SQD group was made by combining ovalbumin injection with Rhubarb purgation, eating and drinking without temperance and overstrains. After the succeed of modeling, divide the AR+SQD group randomly into AR+SQD group and AR+SQD treated with TCM. Before the sdtimulation of part immunization in the nasal cavity, The AR+SQD+TCM group were give TCM treatment (rats given Yiqi Tuoming prescription of 5.58 g/kg/day for seven days). The control group,the AR group,the SQD group and the AR+SQD group were given equal amount of normal saline. Except the normal controls and the SQD group, the other groups were stimulated an hour after giving the drug by 5% ovalbumin solution. The normal controls and the SQD group used the normal saline instead of ovalbumin.From the first day of the modeling, Observe the change of the weight and the common condition on the rats. And especially watch the symptom in the nasal in the period of 30 minutes driping the ovalbumin solution.30 minutes after the last stimulation in the nasal cavity, take the blood from the rats'eye socket, survery the Eosinophils levels, detective the content of interleukine 5 (IL5) and eosinophil cationic protein (ECP) in serum. Execut the animal, strip the maxilla and get the nasal septum mucous membrane, fastened by paraformaldehyde, do the HE staining before pathological morphology. Watch the change of pathomorphism by light microscope. Test the Substance P from nasal septum mucous membrane by Immunohistochemical measurement in the SQD group.Results:The rats in AR and AR+SQD groups were positive for symptoms in the nasal(itch,sneeze,nasal discharge), the EOS in nasal mucus and blood,passive cutaneous anaphylaxis (PCA). The measurements showed increased IL5 and ECP in serum compared with control (P<0.01).There were more EOS and less lymphocyte soaking with vasodilatation and hyperemia in nasal septum mucous membrane. SP rised in all groups, the expression of substance P was increased in AR+SQD animals compared to AR animals. After the treated of TCM, the symptoms of SQD were improved, the symptoms of AR were lightened, increased IL5 and ECP in serum compared with control (P<0.01). The soaking of EOS in nasal septum mucous membrane and expression of substance P were obviouslly lightened.Conclusion:(1)It is feasible that use rhubarb purgation, eating and drinking without temperance and overstrains and ovalbumin as the sensibiligen to build the model of AR+SQD. The animals with SQD easier occur AR than the normal animals. (2) Yiqi Tuoming prescription was effective in treating the rats with AR+SQD. The effection is related to improve inflammation in part of nasal septum mucous membrane and decrease the content of interleukin-5, eosinophil cationic protein, and substance P. This result also reverse proved the succeed of building the model of SQD.
