Toxoplasmosis and brucellosis are two kinds of zoonoses which affect the reproductive health of a variety of mammalian including humans. With the development of social economic and the improvement of people’s living standard, dogs as pets or aiding human work has tightly connected with human life, There is an increasing number of pet dogs in our contry. Therefore, serological surveys in toxopiasmosis and brucellosis of dog are very important for human health. In the present study, canine sera were detested and analysed in several areas of Hunan Province testing and analysis, which can supply us a preliminary understandings about the prevalence of canine toxoplasmosis and brucellosis, incidence of these two diseases and the related factors of infection.A total of 870 canine sera were collected from some big pet hospitals and the Small Animal Protection Associations located in five regions of Hunan Province during 2013 and 2014, and different assays were performed to test the positive ratios of anotibodies against in canine toxoplasma and brucella in these samples.Serological survey of Toxoplasma:Indirect hemagglutination test (IHA) and enzyme-linked immunosorbent assay (ELISA) were used to detect the positive rate of Toxoplasma antibody. The date from these two assays were matched well through comparison, but the sensitivity of the ELISA test is higher than the indirect hemagglutination test. The results from the ELISA assay showed that:positive rates of dog Toxoplasma IgG antibody were 12.46% and 14.97% in 2013 and 2014, respectively, with a slight rise in 2014, The associations of several factors with the T. gondii infection, including gender, age, season, living environment and feeding manners, were analysed in the present study, which suggested that manners of eating and mating may be ciosely related to the indence of canine toxoplasmosis.Serological survey of Brucella:In this study, we used the rose bengal plate agglutination test and ELISA assay to test the Brucella antibodies in canine sera and genotyping was also determined. The date showed that antibody positive rates were 0.54% and 0.80% in 2013 and 2014, respectively. Further confirmation was performed in partial positive samples using rose bengal plate agglutination test, ELISA assay and PCR methods, among them, the result from the ELISA assay was coincided with that detected using PCR, and there may be a false positive detection happended using the rose bengal plate agglutination test. The associations of several factors with the Brucella infection, including gender, age, season, living environment and feeding manners, were also analysed in the this study, the data suggested that manner of mating behavior may be closely related to the indence of canine brucellosis.Based on the above serological survey: no mixed infection with Brucella and Toxoplasma gondii existed in the detected samples. The date also suggest that strengthening management of living and diet of the domestic dogs will be helpful to reduce or even eliminate the indence of canine brucellosis and toxoplasmosis.
