Shaanxi Guanzhong black pig is famous for their excellent characteristics, such as fertility, high meat quality and so on. Thus, Guanzhong black pig is considered to be an excellent dual-purpose breed of meat and fat cultivated specie. With the promotion of the people’s living standard, the meat of black pig become more concerns for customers because of its tasty meat and unique flavors. However, because the pig market environment is declining at the end of last century, the enthusiasm for breeding the Guanzhong black pig of famers had gradually disappeared, this situation led to the decreasing of Guanzhong black pig’s farm scale, along with backward management tool and production technology, the industrialization process of Guanzhong black pig has been seriously constrained. Thus, in order to improve this current situation, three kinds of pig commercial sperm dilutions were selected to test their effects on preservation of Guangzhong black boar sperm in normal temperature, then select the best dilution to explore the effect of deep insemination technology on farrowing performance, and provide test basis for application of deep insemination technology in industrialization process of Guanzhong black pig. The results are as follows:1. After collecting the fresh sperm, sensory tests and microscope tests must be done to detect the performances of sperm, the volume of sperm is 135 ml; the pH value is 7.3; the sperm density is 0.25 billion per ml; the color of sperm is milkiness; the sperm vitality is 0.87 and the malformation rate is 0.16. All the indexes accord with national standard of national standard(GB 23238-2009) and can be used for subsequent tests.2. In this study, the performances of three kinds of pig commercial sperm dilutions were tested by detecting the preservation effect on sperm motility, membrane integrity, acrosome integrity and mitochondrial membrane integrity. Results showed that No. I dilutions had the best preservation effect on Guanzhong black boar sperm for all indexes in normal temperature compared with No. II and No. III dilution(p < 0.05). In addition, the preservation effect of No.II and No. III dilution on sperm motility, acrosome integrity and mitochondrial membrane integrity had no significant differences(p > 0.05). As for membrane integrity, No. III dilution is better than No. II(p < 0.05).3. The effect of different insemination depth on reproductive performanceAll insemination depth, whether 10 cm, 15 cm or 20 cm, had no significant effect on litter size, the number of pigs born alive and the number of pigs born healthy(p > 0.05). Butwith the development of insemination depth, the number of above three indexes also showed an increasing tendency(p > 0.05).4. The effect of different insemination tubes on reproductive performanceAll insemination tubes, whether AMG or Yingtelun, had no significant effect on litter size, the number of pigs born alive and the number of pigs born healthy(p > 0.05).In conclusion, all sperm indexes of Guanzhong Black pig met the criteria, and could be used for experiment. The No. I dilution had the best preservation effect for Guanzhong Black pig’s sperm after a certain time. As for deep insemination, increasing depth of deep insemination can’t improve the reproductive performance of Guanzhong black pig, but with the increasing of deep insemination depth, the reproductive performance have an increasing tendency.
