Effect Of Icariin On The Quality Of Boar Semen Of Guanzhong Black Pig During Normal Temperature Preservation

In recent years,people have become increasingly demanding of meat products,Guanzhong-black pig has received attention again because of its delicious meat quality,and the scale of aquaculture has followed closely,but the application of semen at room temperature and artificial insemination is not standardized.The two major problems of pig semen preservation at room temperature are oxidative damage and bacterial contamination of the mirror during preservation of the semen.According to studies,the extract of Chinese herbal medicine Icariin has free radical scavenging and a certain antibacterial effect.For this purpose,this study tried to add 0 mg/L,5 mg/L,10 mg/L,15 mg/L,20 mg/L,and 25 mg/L icariin to the pig semen diluent BTS,by examining sperm viability,vigor,acrosome integrity,plasma membrane integrity,total antioxidant capacity,and antibacterial efficacy,the feasibility of icariin in replacing gentamicin and its protective effect on sperm were explored.It provides the basis for the study of Guanzhong Black Pig Semen Diluting Powder.This study mainly obtained the following results: 1.When 10 mg/L and 15 mg/L icariin were added to the diluent,the protective effect on sperm viability and vigor was best,and when the dosage was 15 mg/L,sperm acrosome was intact.The protection of sex and plasma membrane integrity is best.Therefore,semen added 15 mg/L icariin had better preservation effect at room temperature.2.Adding different concentrations of icariin in the diluent can improve the anti-oxidant ability of the semen during the preservation of the semen.When the added amount was 15 mg/L,the anti-oxidation ability of the semen was the strongest,and the oxidative damage to the spermatozoa was even more reduced.When the amount was 10 mg/L,the semen had good antioxidant capacity.3.Icariin has a certain antibacterial effect against Escherichia coli and Staphylococcus aureus in in vitro antibacterial tests;icariin is added to seminal diluent,Can inhibit the growth of bacteria in semen to a certain extent.4.The combination of icariin with gentamycin has a good antibacterial effect,15 mg/L icariin can replace 75% of gentamycin,and is similar to the standard addition amount of gentamicin.The antibacterial effect and semen preservation effect at room temperature,and when 15 mg/L icariin replaces 50% gentamycin,its antibacterial effect is similar to the standard addition amount of gentamicin,but its semen is stored at room temperature.The effect is better than the standard addition of gentamicin.In conclusion,the addition of 15 mg/L icariin to BTS has the best protective effect on sperm preservation at room temperature,and also has the best antioxidant effect;icariin has both E.coli and S.aureus.A certain antibacterial effect,when 15 mg/L icariin was used instead of 50% gentamicin,its antibacterial effect was similar to the standard addition amount of gentamycin,but the preservation effect of semen at room temperature was higher than the standard addition amount.Gentamicin is better.
