| The principal components of seminal plasma are proteins,enzymes,lipids,sugar and prostaglandins,inorganic ions,and cytokines from the accessory gonadal glands,which protect the sperm during its preservation.The dilution of semen has been collected based on the sperm count,ignoring the effect of seminal plasma on sperm protection,and also neglecting the regulation of seminal plasma on the reproductive system of the female animal.In this study,semen is diluted by adding different concentrations of semen for normal temperature preservation.Firstly,this experiment detects regular biochemical indexes and antioxidation ability of seminal plasma and provides a viable basis for seminal plasma preservation at room temperature.Secondly,this experiment studies the effects of different concentrations on the quality index,antioxidation ability and energy metabolism of semen at room temperature.The optimum concentration of seminal plasma is detected and screened.Finally,the seminal plasma’s deleterious effect on seminal fluid is explored.The effect of seminal plasma on the preservation of semen at room temperature is investigated after bacterial growth suffered as a result of the addition of antibacterial agents.The main results obtained in this study are as follows:1.Seminal plasma contains functional substances such as proteins,trace elements,and macromolecules.Antioxidation ability of seminal plasma increases with the increasing concentration and the total accommodative capacity of seminal plasma in Guanzhong-black pigs is higher than that of Yorkshire.The addition of seminal plasma may have a beneficial effect on the preservation of semen at room temperature.2.Adding seminal plasma at a high concentration of 50% and 75% can significantly reduce sperm viability and the integrity of the acrosome and plasma membrane(P < 0.05);low concentrations of seminal plasma can improve the quality of semen stored at room temperature.Among them,25% seminal plasma concentration had the best effect(P < 0.05).3.Adding 25%,50% and 75% seminal plasma can significantly increase the total antioxidant capacity of semen,of which 50% of the total antioxidant capacity is highest(P < 0.05);25% seminal plasma can significantly reduce semen C2 Aldehyde content(P < 0.05);15% seminal plasma can significantly reduce semen’s reactive oxygen species(P < 0.05).4.The addition of seminal plasma can increase the content of ATP in semen,but only 75% of seminal plasma is significantly different from the control group;addition of 25% seminal plasma can significantly improve sperm phosphofructokinase(PKF)and malate dehydrogenase(MDH)activity.5.The content of bacteria in semen is positively related to the concentration of seminal plasma.After the combination of seminal plasma and antibacterial agent to inhibit the bacterial growth of semen,seminal plasma can substantially increase the viability and viability of sperm,and 50% of the sperm are added.The pulp effect is best(P < 0.05).In summary,there are numerous kinds of substances beneficial of the semen preservation in Guanzhong-black pig seminal plasma.The addition of seminal plasma can increase the quality of semen stored at room temperature,and 25% of the seminal plasma can drastically improve the sperm performance,membrane integrity,Antioxidant capacity and energy metabolism levels.However,seminal plasma increases the content of bacteria in semen,and a better preservation effect can be obtained by adding antibacterial agents in combination. |
