| Artificial insemination technology is widely utilized in the practice of pig production,and sperm insemination equipment is a significant aspect influencing the effect of artificial insemination.Wuhan Tianchu Biotechnology Co.,Ltd.’s new type of insemination catheter is the market’s first insemination equipment that integrates sperm storage bag and insemination catheter all in one.It is also a bionic design that resembles natural boar mating.The semen can naturally flow into the sow’s uterus under the pressure of the elastic silicone membrane of the novel type of insemination catheter and the cervical lining.In comparison to the traditional plastic semen storage bottle,the new type of insemination catheter with an elastic silicone membrane will apply pressure to the stored semen.Due to a lack of research on the effect of pressure on the quality of sperm preservation,the experiment collected sperm from Yorkshire and Duroc,diluted and filled it into new type insemination catheter and conventional plastic sperm storage bottles,and measured the internal pressure of the two preservation devices.Sperm motility,deformity rate,sperm acrosome integrity rate,plasma membrane integrity rate,and mitochondrial membrane potential were all measured simultaneously while stored at 17°C for 0 h、24 h、48 h、72 h、96 h、120 h and 148 h.Indicators and measuring standards are used to relate to "Semen Production"(GB23238-2009).According to the findings of the tests,180 healthy 、 2-4 parity sows with no reproductive problems were used in artificial insemination and breeding trials utilizing a new type of insemination catheter and traditional plastic semen storage bottles.Differences in variables like conception rate and litter size were documented and statistically assessed.The following are the key findings:1.From 0 to 48 hours after semen storage,the sperm motility of the two boars stored in the new type of insemination catheter and standard sperm storage bottles was not significantly different(P>0.05).Sperm motility in the new type of insemination catheter differed significantly from that in the usual sperm storage bottle after 48 hours of storage in two pig breeds(P<0.05).2.The CASA system detected the motility parameters of Yarkshire and Duroc sperm in the new type of insemination catheter and standard sperm storage bottles between 0and 144 hours of semen storage.The sperm motility index of the two versions was not substantially different between the new type of insemination catheter and the conventional sperm storage flask between 0 and 24 hours of semen storage(P>0.05);The VSL of the two breeds of pig sperm in the new type of insemination catheter and standard sperm storage bottles was considerably different between 24 and 48 hours of semen storage(P<0.05);There were substantial variations in the new type of insemination catheter and the standard sperm storage bottle group after 48 hours of storage between the two breeds of pig sperm VAP(P<0.05);The four indexes of VSL、VAP、VCL and ALH of the two breeds of pig sperm in the new type of insemination catheter were significantly lower after 96 hours of storage than those in the standard sperm storage bottle group(P<0.05).3.After storing the sperm for 96 hours,the new type of insemination catheter had greater rates of sperm abnormalities than standard vials(P<0.05);The acrosome integrity rate of the two breeds of boar sperm in the new type of insemination catheter was considerably different from that in the conventional sperm storage bottle after 72 hours of storage(P<0.05);The mitochondrial membrane potential of the spermatozoa of the two types of semen stored in the new type of insemination catheter was decreased after 72 hours of storage compared to the conventional semen storage bottle(P<0.05).4.The boar semen saved within 48 hours was chosen based on a study of the semen test results stored in the two insemination devices,and the new type of insemination catheter and traditional plastic semen storage bottles were utilized for breeding testing.The conception rate of sows with artificial insemination using the novel type of insemination catheter had a conception rate of 69 percent,with an average litter size of13.29,and the conception rate of sows with conventional insemination vials was 72 percent,with an average litter size of 13.81.There was no significant difference between groups(P>0.05)。 Furthermore,there was no difference between the new form of insemination catheter insemination and the conventional insemination bottle in terms of the number of healthy piglets per litter,the number of stillbirths per litter,or the birth litter weight.artificial fertilization(P>0.05).However,when the breeding effectiveness of the two insemination devices is compared,the number of sows bred by six breeding staff is 90.The new form of insemination catheter takes 84 minutes to use and has a efficiency per capita of 10.7 heads/hour.The traditional sperm storage bottle takes 220 minutes to use,and the efficiency per capita is only 4.16 heads/hour.In conclusion,the semen maintained in the new type of insemination catheter may sustain more than 60% sperm motility within 72 hours,which is consistent with artificial insemination standards.Simultaneously,artificial insemination and breeding trials reveal that the new type of insemination catheter and traditional sperm storage bottles have no significant effect on sow conception rate and litter size.However,the new type of insemination catheter’s autonomous insemination technology can considerably cut breeding time,improve breeding efficiency,and reduce labor costs.As a result,the findings of this experiment demonstrate that the new type of insemination catheter is suitable for large-scale promotion,although the effect of breeding must be tracked and examined further. |
