| Cow vagina flora plays a very important role in maintaining normal physiological function of the reproductive tract and preventing of disease. At present, although there are numerous research reports of cow vaginal flora, but we still know little of the diversity and dynamic changes of vaginal flora. This experiment is in order to further study the cow vaginal flora and complex changes, carried on the analysis and comparison of the use of high throughput sequencing method. The experiment selects Holstein cows in Changchun City area as experimental animals, collecting three healthy cows and three cows suffering from endometritis postpartum 6-8d and 34-36 d vaginal mucus. By extracting the sample amplification of bacterial total DNA, 16 S rDNA V3 fragment and establishment of high throughput sequencing library, then sequencing of high flux sequencing library and finally through the biological comparison of health and endometritis cows postpartum vaginal bacteria on different days of the swarm diversity differences.In this experiment, a sample of high-throughput(Illumina MiSeq) sequencing, after data processing, finally get 710,382 effective sequences, and then by bioinformatics analysis showed 165 Species 20 Phylum, there are some unknown bacteria classes, the number of bacteria in each sample, and the abundance of species in the species level classification are different. Firmicutes as healthy cows and suffering from different time periods within the uterus vaginal postpartum endometritis cows shared door dominant bacteria, Actinobacteria for the above two cows postpartum vaginal advantage 34-36 d bacteria door, Fusobacteria for 6-8d sick cows postpartum vaginal dominant bacteria door. The study shows the construction of the experimental cows vaginal flora is a dynamic process of evolution, the complexity of the different stages of postpartum endometritis healthy cows and cows suffering from vaginal flora is increasing with the extension of post-natal period and found that the number and types of bacteria in the vagina healthy cows are the most in postpartum 34-36 d.Generally speaking, the experiment proves that in the vagina of cow has its unique flora structure, finds out the differences in the vagina of healthy cows and endometritis cows in bacterial community structure, and dramatically changed over time. |
