The Difference Of Vaginal Bacterial Diversity Between Healthy And Endometritis Sow And Screening The Lactobacillus

The sow endometritis is a kind of common diseases of genital organs in postpartum as a result of bacterial in faction,it leads to reproductive disease and has great impact on the sow industry.In this study,the healthy and endometritis sow's vaginal mucus were used as research objects,carried out on the analysis and comparison of the use of high throughput sequencing method.In addition,this paper isolated several strains of lactic acid bacteria from the healthy sow's vaginal swab,and tested the probiotic function of bacteria static ability,produced acid,hydrogen peroxide production for this strains,so that selected excellent performance of lactic acid bacteria.In this experiment,the result of high-throughput(Illumina MiSeq)sequences showed 245,882 effective sequences from all samples,and by bioinformatics analysis showed 9 phylum and 33 genus.Furthermore,there are some unknown bacteria classes.In each sample,varieties of abundance in the species level classification are different.The vaginal mucus of healthy sow was dominated by Firmicutes,in the next place dominated by Proteobacteria and Bacteroidetes.However,the dominated phylum in endometritis was Proteobacteria,followed by Firmicutes,Bacteroidetes,Fusobacteria.At the genus level,the microbial species in endometritis sow's vaginal mucus are richer than healthy sow.And compared with healthy sow sample,the bacteria genus with a significant difference are Escherichia-Shigella,Fusobacterium,Clostridium,Aquabacterium,Staphylococcus,Prevotellaceae,Porphyromonas.Based on traditional cultivation methods,the test isolated 2 Lactobacillus johnsonii,2 Lactobacillus salivarius,1 Lactobacillus murinus,2 Pediococcus acidilactic and 1 Lactobacillus animals from the healthy sow's vaginal swab.These lactobacilli have good live features.The research indicates the differences of bacterial diversity in vaginal of healthy and endometritis sow by high-throughput(Illumina MiSeq)sequences,and obtains 8 strains of Lactobacillus.The result not only provides the theoretical support for vaginal bacterial diversity of sow,but also provides reference for the development of micro ecological probiotics.
