Meat safety has been paid more and more attention and traceability becomes necessary. There are many traceability methods for meat products, such as label traceability, isotope traceability and DNA traceability. Compared with other methods, DNA traceability is easier to genotype and more suitalbe for large-scale automatic detection. It can not only be used to trace for live animals, but also for meat products. The SNPs marker, one kind of the DNA markers, is easier to genotype and suitable for rapid, high-throughput detection. Therefore, in this study, a new method for DNA traceability of pork was investigated.1. A new method for SNPs markerA comparison of high resolution melting analysis (HRM) was performed with RFLP-PCR analysis and TaqMan probe analysis in terms of accuracy, ease of operation, the costs of detection and so on. The results show that high resolution melting analysis is more accurate, cheaper and more suitable for large-scale automatic analysis for DNA traceability, especially screening SNPs markers.2. Comparison of DNA extraction from different tissuesThe DNA extraction and detection effiency was compared among muscle, blood and hair from the same animals using HRM. The results showed that all the three tissues are suitable for high-resolution melting analysis. However, the blood and muscle DNA samples have better repeatability.3. The screening of effective SNPs markersTen pig SNPs markers were designed according to the reported references or NCBI database.299 samples were used to analyze the allele heterozygosity of these SNPs markers. The results showed that the allele heterozygosities of all the 10 SNPs markers were above 0.4. Therefore, all of the 10 SNPs markers can be used for pork DNA traceability.4. The framework of DNA traceability systemA traceability system combined RFID with DNA were developed. SNPs coding was designed for the system.
