Intradermal test, prick test and patch test, which are commonly used in clinical skin test, not only could largely avoid serious drug allergies but also provide necessary diagnostic information for allergic diseases. There are the following problems for intradermal test and skin prick test:the obvious pain, the skin damage the risk of infection from skin injury; the overdose risk in the intradermal test which can cause shock or even death during the test; the longer time for patch test and the obvious pain from removing the patch.Over these years, researchers have been exploring non-invasive, fast, convenient and painless skin test methods such as laser shock transdermal. This method is non-invasive transdermal administration of trace, the dose of which is far less than the intrademal injection, and it is fast and easily-operated. However, present research is not sufficient, such as, there’s no explicit report of how to control the impact or what effects of this method will have.To improve the efficiency of the laser shock transdermal administration, the paper used self-developed thin shock wave energy conversion laser device to operate the experimental tests. It was used to research what the influence of the laser energy value(mJ), the defocusing amount and the cartridge diameter of liquid storage box on the laser impact. It was proved that while there was a smaller defocusing amount, the force generated by the laser shock showed the "saturated" feature with the increase of single-pulse laser energy; while there was a larger defocusing amount, the force generated by laser shock showed a "fluctuated" feature with the increase of laser pulse energy. In practice, the "saturated" feature was useful for adjusting the laser energy to control the impact force to achieve the purpose of transdermal administration without damaging the skin. The "fluctuated" feature also indicates that larger defocusing amount was not conducive to control impact.In the paper, a series of skin tests were conducted by the converting device with specific parameters. The histamine which is the standard reference solution of positive skin test was used. The experimental results showed that with smaller defocusing amount,the intensity of skin erythema reaction was also presented the "saturated" feature following the increase of the laser pulse energy. This result was consistent with the trend of the impact of the laser.The results also showed that, the effects of laser shock wave on the transdermal histamine solution reaction at least contained two aspects:one is the compression impact of shock wave. The second is the temporary permeability was formed on the skin surface by laser impact. The skin also showed a positive reaction and the subsequent role of the share was more transparent pre-impact extrusion effect.Results of this study can provide basic research basis for the clinical practice applications of laser shock test.
