Stone Balsam Promote Wound Finger To Explore The Mechanism Of Defect

Purpose:To observe the influence Chinese medicine Shi Xiang Cream has on the model of defect traumatic soft tissues of domestic rabbits,referring to the time of wound healing,healing rate,the proliferating condition of the fibroblast inside of the wound granulation tissues,as well as the impact of b-FGF and TGF-β1.Meanwhile disscussing the mechanism of how external application of Chinese medicine could promote the repair of soft tissues,respectively from morphologic,cellular and molecular level.Method:Divide30adult male rabbits randomly into GroupA,B and C,each group has10and Group A is Shi Xiang Cream,Group B is rb-bFGF and Group C is control group.Firstly,i employ the typical molding of full-thickness skin concis,using Pentobarbital to do abdominal cavity anesthesia.As soon as it works,i do the depilation of the back of the rabbit using8%sodium sulphide,then sterilize in the common practice and resect overall width of cortex,do a2×3square centimetre cut and wipe out the errhysis.Lastly,i handle the surface of the wound respectively with external application of Shi Xiang Cream in Group A,external spray rb-bFGF in Group B and cover with surgical dressing,no special handling in Group C but covering wounds with the clean surgical dressing.Afterwards,i do reput on the medicine respectively in day3,6,9and12after surgery,and observe the healing condition of the wound,then i collect the newly-born granulation tissue and the tissue of the wound incisions to do paraffin embedding,section fixing and HE staining. Lastly,i observe cell proliferation of the fibroblast and macrophage with the help of high power lens,meanwhile,i detect the expression of protein b-FGF and TGF-β1using SABC method under400-times light microscope and do the analysis BI-2000using the image analysis system,showed by average gray value.Consequence:Group Shi Xiang Cream and Group rb-bFGF regrow in advance compared to Control Group,and the healing cycle’s shorter,the rate of wound healing in the same period’s higher,last but not least,the occurence of epithelial crawling’s sooner.The group Shi Xiang Cream and Group rb-bFGF show no obvious differnence after statistics,in the meanwhile matters to the Control Group in the statistics level.Under the same condition of samples,the expression of protein b-FGF in Group3shows difference after6days with trauma(P<0.05),the Group Shi Xiang Cream and Group rb-bFGF are averagely higher than the Control Group,it matters in statistics.The expression of protein TGF-β1shows difference after6and9days with trauma(P<0.05),the Group Shi Xiang Cream and Group rb-bFGF are averagely higher than the Control Group,it matters in statistics.Meantime,it shows difference between groups in the growth status of granulation tissue in the same samples in period,and difference within groups in the conditon of different times.The expression of protein TGF-β1goes higher as time goes by.Conclusion:The Chinese medicine Shi Xiang Cream can accelerate the cell proliferation of fibroblast and macrophage,it works in the way of stimulating the cells in the wounds and therefore results in the production of endogenous growth factors TGF-β1and b-FGF,naturally arousing the cell proliferation of fibroblast which in turn promoting the repairment of the wounds.
