Purpose:To investigate the mechanism of white mange medicine mixture on the mousemodels of psoriasis and to observe its effect s on keratinocyte expression ofproliferating cell nuclear antigen,T lymphocytes apoptosis and serum chemokineGM-CSF,and to explore its possible mechanism.Material and method:1.AnimalKunming female mice,Mice aged6to8weeks,Each body weight18-22g,A totalof84(the experimental animal center of Liaoning University of TraditionalChinese Medicine).2.Methods(1) Animal grouping and treatment84female Kunming mice,Divided into seven groups by random numbertable,Using a random number table,84mice were randomLy divided into7groups:blank control group,consumpition of silver particles group,avia A group, negtivecontrol group,high dose of white mange mixture group,middle dose of white mangemixture group,low dose of white mange mixture group.Dosage Adult100g/70kg daily,Mice administered dose to be converted inaccordance with the human and mouse proportion of body surface area,Calculatedin high,middle and low dose group,the concentrations of gastric lavage were1g/mL,0.5g/mL,0.25g/mL,in negtive control group was0.2mg/mL,in avi A groupwas0.25mg/mL.Blank control group normal feeding.Executing for28days.(2) Model of psoriasis preparationIn addition to the72mice (outside blank control group),intraperitoneal injection of estradiol benzoate injection of0.2mg/10g,once daily,for3consecutive days,all mice were in estrogen period.Each groups of animals wereweighted respectively,according to the weight of a perfusion feeder intragastricadministration,once a day,for4weeks.Every two days weighted1time to fillthe medicine dose adjustment.(3) Sample collection and detection(a)Inhibitory effect of S-P was observed by immunohistochemistry,theproliferation of murine KC cell nuclear antigen PCNA.(b) Electron microscopy spleen T lymphocyte apoptosis in mice.(c) The production of GM-CSF was detected by ELISA.Results:1. Vaginal epithelial keratinocytes detection of proliferating cell nuclearantigen PCNA-positive cells.Compared with consumpition of silver particles group,white mange mixture inhigh,medium,low dose groups induced apoptosis of T lymphocytes,inhibition ofKC proliferating cell nuclear antigen PCNA,SP immunohistochemical staining wasobserved in control mice,the expression of PCNA-positive rate and coloringdepth,slightly stained,clear hierarchy,deep coloring of model mice,the Herbalhigh dose,medium dose,low-dose group compared with the model are coloreddeep,most evident in the high dose group,negtive control and avi a group coloringdarker,clearer hierarchy.2. The results of spleen T lymphocyte apoptosisionCompared with blank control group,the white mange mixture of high,medium andlow groups can apoptosis,electron microscopy the normal group visibilitylymphocytes nuclear large round,distribution of heterochromatin in the nuclearperiphery,prominent nucleoli,chinese medicine high-dose,medium-dose,low-dosegroup than blank apoptotic bodies,with high doses of the most obvious,avia Aand negtive control group were seen few apoptotic bodies.3. The results of ELISA in serum GM-CSF content Compared with the control group, the serum GM-CSF content in each model weresignificantly increased (P <0.01); compared with the negative control group,Baibi mixture in each dose group decreased the content of GM-CSF (P<0.01);andcompared with negative control group,white mange mixture in each dose groupdecreased(P<0.01),but high dose group with positive control the differencebetween is not obvious(P>0.05).Conclusion:1.White mange mixture could significantly inhibit KC proliferating cell nuclearantigen PCNA of mice model of psoriasis.2.White mange mixture can induce the apoptosision of spleen T lymphocytes ofmice model of psoriasis.3.White mange mixture can reduce the expression of serum GM-CSF.4.White mange mixture may play a role in the treatment of psoriasis effect byinducing T lymphocyte apoptosision,inhibition of epithelial KC proliferatingcell nuclear antigen PCNA,and decreasing the generation of serum GM-CSF....
