ObjectiveTo investigate the mechanism of white mange medicine mixture on the mousemodels of psoriasis and to observe its effect s on mitosis index、 T lymphocytesproliferation and serum chemokine IL-8,for its wide application in clinicalprovide theoretical and experimental evidence.Methods1.AnimalKunming mice, female, body weight18-22g, purchased from the experimentalanimal center of Liaoning University of Traditional Chinese Medicine.2. Methods(1) Animal grouping and treatmentUsing a random number table,84mice were randomLy divided into7groups:blank control group, A via A group,consumpition of silver particles group,negtive control group, high dose of white mange mixture group, middle doseof white mange mixture group, low dose of white mange mixture group.In high, middle and low dose group, the concentrations of gastric lavagewere1g/mL,0.5g/mL,0.25g/mL, in avi A group was0.25mg/mL,consumpition of silver particles group was0.053g/mL. Rats in negative controlgroup daily with an equal volume of normal saline lavage, blank control groupdid not exert any intervening factors. Above each medication were executionfor28days.In addition to the72mice (outside the normal group), intraperitonealinjection of estradiol benzoate injection of0.2mg/20g, once daily, for3consecutive days, all mice were in estrogen period. Each groups of animals were weighted respectively, according to the weight of a perfusion feederintragastric administration, once a day, for4weeks. Every two days weighted1time to fill the medicine dose adjustment.(3)Sample collection and detection(a) The effect of white mange medicine mixture on mitosis exponential of vaginalepithelial cells was measured using HE staining.(b) The proliferation of mice spleen T lymphocytes was detected by MTT.(c) The production of IL-8was detected by ELISA.Results1.The results of vaginal epithelial cell mitotic indexCompared with negative control group, white mange mixture in high, medium,low dose groups were significantly decreased estrogen mouse vaginal epithelialcell mitotic index, but high dose group with positive control the differencebetween is not obvious (P>0.05). Observed under the light microscope, blankcontrol group in mouse vaginal epithelium of normal squamous epithelium, thelevel of clear, negative control group of mouse vaginal epithelial basal cellmitotic cells more, Chinese medicine high, medium, low dose groups andpositive control group than those in negative control group of basal cell mitoticcounts were reduced, and high dose group was most obvious in Chinese medicinegroup.2.The results of spleen T lymphocyte proliferationCompared with blank control group, T lymphocytes have different degreeincreased in each of the model groups, but the difference was not statisticallysignificant (except model group with)(P>0.05); compared with negative controlgroup, T lymphocyte proliferation was significantly suppressed in each dosegroup of white mange mixture(P <0.01), but high, medium dose groups withpositive control and between groups the difference between is not obvious(P>0.05).3.The results of ELISA in serum IL-8content Compared with blank control group, the content of IL-8in serum of the modelgroups was significantly increased (P <0.01); and compared with negative controlgroup, white mange mixture in each dose group decreased (P <0.01),but highdose group with positive control the difference between is not obvious (P>0.05).Conclusion1.White mange mixture could significantly inhibit the vaginal epithelial mitosisof mice model of psoriasis.2.White mange mixture can inhibit the proliferation of spleen T lymphocytes ofmice model of psoriasis.3.White mange mixture can reduce the expression of serum IL-84.White mange mixture may play a role in the treatment of psoriasis effect byinhibiting T lymphocyte proliferation,epithelial cell mitosis,and decreasingthe generation of serum IL-8....
