Objective:To explore the Safety and Effects of Cholinesterase inhibitor combined with glutamate receptor antagonists, lipid-lowering drugs triple therapy on Vascular Dementia.Methods:This study was a randomized and controlled clinical trial. Patients came from Rehabilitation Center of Psychiatric Hospital in Xiangxi Autonomous Prefecture, Hunan. Sixty patients of Vascular dementia who were admitted into the trail were randomly divided into two groups. The treatment group (n=30) received the triple therapy (Donepezil+Memantine+Atorvastatin), and the control group (n=30) received Donepezil. The treatment lasted for6months. Before and after the6-month treatment, patients from two groups were assessed by mini-mental state examination (MMSE), Blessed-Roth dementia scale (BRDS) and activity of daily living scale (ADL). Treatment emergent symptom scale (TESS) and other relevant laboratory detections (blood, urine and stool tests, electrocardiogram, hepatorenal function and electrolyte) respectively to evaluate drug safety.Results:MMSE scores in treatment group were better than those in control group (P<0.05). ADL and BRDS scores were worse in treatment group (P<0.05). The total effective rate in treatment group was higher than that in control group (83.3%and53.3%),(χ2=6.239, P<0.05).Both group showed no significant differences in TESS scores (P>0.05)Conclusion:Combined use of cholinesterase inhibitors, glutamate receptor antagonist and reducing blood fat drugs is an effective method to treat vascular dementia, and this combined use would not affect the safety.
