Objective:According to the requirements of "Technical Specifications for Health Food Inspection and Evaluation"(2003edition), the stability and the functions of lowering blood lipid and improving the immunity of peony seed oil soft capsule were investigated so as to provide a scientific basis for developing peony seed oil soft capsule as a health food.Methods:1. The study on the stability:Three successive batches of peony seed oil soft capsules (20121015,20121016,20121017) were placed in a humidity chamber with relative humidity of75%and temperature of38±1℃. Twenty three indicators, such as the contents of linolenic acid and linoleic acid, the residues of lead, arsenic, benzenehexachloride (BHC) and mercury, the acid value, peroxide value, microbiology testing and so on, were examined respectively at the0,1,2,3month to evaluate the stability of the products in three months.2. The study on the blood lipid lowering functionsregulation:Fifty healthy adult rats were divided into five groups. They were control group (feeding normal diet and corn oil), model control group (feeding high-fat diet and plus corn oil), low-dose group (feeding high-fat diet plus peony seed oil (500mg/kg, i.g, equivalent to5times of the recommended amount for human)), middle-dose group (feeding high fat-diet plus peony seed oil (1000mg/kg, i.g, equivalent to10times of the recommended amount for human)), and high-dose group (feeding high fat-diet plus peony seed oil (3000mg/kg, i.g, equivalent to30times of the recommended amount for human)). All rats were continuously given the test sample for36days. The serum levels of TC, TG, LDL-C, and HDL-C were determined with no fasting at the end of the experiment.3. The study on the immunity improving functionsion:Two hundred healthy SPF Kunming mice were divided into four immune groups. The first immune group (SOfemale mice) was used for the delayed type hypersensitivity (DTH) test, and the determinations of serum hemolysin and the number of antibody-forming cells. The second immune group (50female mice) was used for mouse carbon clearance test. The third immune group (male mice50) was used for testing the phagocytosis of mouse peritoneal macrophages on chicken erythrocyte. The forth immune group (male mice50) was used for mouse lymphocyte transformation test and activity determination of mouse NK cells. Each group above was divided into five small groups which were water control group, solvent control group, low-dose group, middle-dose group, and high-dose group, and10mice in each small group. The doses of low-dose group, middle-dose group, and high-dose group were0.5g/kg,1.0g/kg, and3.0g/kg. Mice were gavaged once a day with a volume of10ml/kg) and continuously fed for30days.Results:1. The results of the studies on stability showed that the peony seed oil soft capsules stored under38±1℃and the relative humidity of75%for3months were stable as indicated by inolenic acid and linoleic acid contents, acid value, peroxide value, and disintegration time.2. The results of the studies on blood lipid lowering functions showed that the TC, TG, and LDL-C levels of the model control group were significantly increased compared with the control group, and the difference had statistical significance (P<0.01), which indicated the high-fat model was established sucessfully. The TC, TG, and LDL-C levels of the high-dose group were significantly decreased compared with the model control group, and the difference had statistical significance(P<0.05). The HDL-C level of high-dose group was significantly increased, and the difference had statistical significance (P<0.01).3. The results of the studies on the immunity improving functions showed that the different doses of peony seed oil soft capsule were taken orally by mice for30days, and there were no effects on body weight and spleen and thymus indices. The phagocytosis of mouse monocyte-macrophages were compared between the middle, high-dose group and water control group, and the difference had statistical significance (P<0.05). The numbers of antibody-producing cells of mice were compared between the middle, high-dose group and water control group in the test of the humoral immune function, and the difference had statistical significance (P<0.05). The serum antibodies of mice of the middle, high-dose group were significantly increased compared with water control group (P<0.05). The peony seed oil soft capsules had no effects on cellular immune function of mice and the activity of NK cells.Conclusions:1. The peony seed oil soft capsules stored under the conditions of relative humidity of75%and the temperature of38±1℃for3months was stable. It was in line with the "National Health Food Standards" issued by the the Ministry of Public Health.2. The results of experiments showed that peony seed oil soft capsules had blood lipi lowering functions.3. The results of experiments showed that peony seed oil soft capsules could enhance the immune function.
