The Antioxidant Function Research Of Eucommia Seed Oil And Preparation Of Its Realated Compound Preparation

The aim of this paper is to study the antioxidant function of eucommia seed oil and the preparation processes,the quality control standards,the stability and the physiological activity of eucommia ulmodies natto soft capsule.Specific results were as follows: 1 The antioxidant function research of eucommia seed oilThe crude eucommia seed oil was from the samara of eucommia ulmodies by using supercritical carbon dioxide extraction method.Then the refined eucommia seed oil obtained after refining.Eucommia seed oil was up to edible product standard through determination.The adult male mice were given 200mg/(kg·BW),400mg/(kg·BW)and 1.2g/(kg·BW)dose by gavaging once a day.Then poisoned mice by bromobenzene after gavaging for continuous 30 days.Detemined the antioxidant function indexes in blood and liver of mice.Low dose groups of eucommia seed oil could significantly increase T-AOC(p<0.05),GSH-Px and SOD activity(p<0.01),and obviously decrese MDA content(p<0.05).The result showed that eucommia seed oil has the antioxidant function.2 The preparation technology research of eucommia ulmodies natto soft capsuleEucommia ulmoides natto soft capsule content consists of eucommia seed oil,flaxseed oil,natto dehydro freezing powder,superfine powder of eucommia ulmoides male flower,beeswax and natural vitamin E.It is mainly used for hyperlipidemia.The dosage of suspending agent beeswax in the soft capsule contents was 2%.The stability of suspension system can maintain good.Capsule shells main component percentage was:gelatin,glycerin,water=1:0.4:1.The dosage of titanium dioxide in the capsule shell was 0.7%,and methyparaben was 0.02%.Preparation technology:Mixed eucommia seed oil,flaxseed oil and beeswax at 60~70?.Added vitamin E when the temperature dropped to 35? and set aside.Then mixed eucommia ulmodies male flower superfine powder and natto dehydro freezing powder,and sifted through 100 mesh sieve.Then added it to the oil phase,blending,filtration,potting and made it into soft capsule.3 The quality control standards research of eucommia ulmodies natto soft capsuleThe limit value of each indicators had been determined and formulated,based on Chinese Pharmacopoeia(2015).The content of ?-linolenic acid were determined by GC.The average content of ?-linolenic acid was 0.15 g per grain in soft capsule.The methodological study showed that the linearity,precision,reproducibility,stability and recovery were all satisfactory.The content of ?-linolenic acid was not less than 0.13 g per grain in eucommia ulmodies soft capsule.4 The preliminary stability research of eucommia ulmodies natto soft capsuleWe studied the preliminary stability under normal temperature conditions,and the result showed that the stability was good among 3 months.5 The pharmacodynamics and toxicology research of eucommia ulmodies natto soft capsuleWe studied the acute toxicity and hypolipidemic experiment.In the hypolipidemic efficiency experiment,used the content of soft capsule to gavage mice by the recommended doses of 167mg/(kg·BW),333mg/(kg·BW)and 1g/(kg·BW)for treatment.The result showed that the soft capsule contents could significantly reduced TG and TC content in mice plasma(p<0.05),and also could reduced LDL-C content(p<0.01).increased HDL-C content(p<0.01).It also could control weight gain of high-fat model mice(p<0.01),and also could improve lipid metabolic disorder of mice liver coefficient change(p<0.05).The study showed that eucommia seed oil has the function of antioxidant,and could be protective.The compound preparation eucommia ulmodies natto soft capsule preparation technology was reasonable,the quality could be control and stability was good,has the hypolipidemic activity,less side effects than western medicine.
