Objective: The expression of PKC, Annexin A2and S100A10proteins, whichthey were reported the differential expression proteins of gastric cancer, was detectedin different gastric tissue. The clinical significance of them expression was analyzed,and will provide valuable data for clinical early diagnosis, clinical molecularpathology type and evaluation the clinical result of gastric cancer patients.Methods: We respectively collected the normal gastric mucosa, atypicalhyperplasia, gastric cancer and lymphonode metastatic carcinoma tissue and extractedthe total protein of above-mentioned different tissue. The expression of PKCα,Annexin A2and S100A10proteins was checked by Western-blot in different gastrictissue. Moreover, the expression of PKCα, Annexin A2and S100A10proteins wasanalyzed by immunohistochemistry in tissue array included of normal gastric mucosa,atypical hyperplasia, gastric cancer and lymphonode metastatic carcinoma tissue. Theclinical pathology significance of their expression was evaluated in gastric cancertissue.Results:1. The Western-blot result showed that the expression of PKCα, AnnexinA2and S100A10proteins was higher in gastric cancer and lymphonode metastaticcarcinoma tissue than in normal gastric mucosa and atypical hyperplasia tissue(P<0.01). Moreover, their expression was higher in atypical hyperplasia tissue than innormal gastric mucosa (P<0.01).2. The immunohistostaining consequence exhibitedthat the positive expression rate of PKCα protein was respectively8.82%(3/34),34.62%(9/26),76.54%(62/81) and78.18%(43/55) in normal gastric mucosa,atypical hyperplasia, gastric cancer and lymphonode metastatic carcinoma tissue. Its positive expression rates were respectively60%(3/5),70.59%(12/17) and79.66%(47/59) in gastric well, middle and poorly differentiated adenocarcinoma. The positiveexpression rate of Annexin A2protein was respectively5.88%(2/34),30.77%(8/26),79.01%(64/81) and76.36%(42/55) in normal gastric mucosa, atypical hyperplasia,gastric cancer and lymphonode metastatic carcinoma tissue. Its positive expressionrates were respectively60%(3/5),76.47%(13/17) and81.36%(48/59) in gastric well,middle and poorly differentiated adenocarcinoma. The positive expression rate ofS100A10protein was respectively2.94%(1/34),19.23%(5/26),59.26%(48/81) and63.64%(35/55) in normal gastric mucosa, atypical hyperplasia, gastric cancer andlymphonode metastatic carcinoma tissue. Its positive expression rates wererespectively40%(2/5),52.94%(9/17) and62.71%(37/59) in gastric well, middle andpoorly differentiated adenocarcinoma.3.The positive expressin rate of PKCα,Annexin A2and S100A10proteins was higher in gastric cancer and lymphonodemetastatic carcinoma tissue than in normal gastric mucosa and atypical hyperplasiatissue (P<0.01). Their expression was higher in atypical hyperplasia tissue than innormal gastric mucosa (P<0.01). The expression of PKCα, Annexin A2and S100A10proteins was correlated with tissue differentiation level and their positive expressionrates were high in gastric lower differentiated adenocarcinoma (P<0.01).Conclusion:1. The expression of PKCα, Annexin A2and S100A10proteins was up-regualtion ingastric cancer and lymphonode metastatic carcinoma tissue compared with innormal gastric mucosa and atypical hyperplasia tissue. And their expressions werehigher in atypical hyperplasia tissue than in normal gastric mucosa tissue.2. The expression of PKCα, Annexin A2and S100A10proteins was related withoccurrence, differentaiation level of tissue and lymphaden metastasis of gastriccancer.
